MicroRNA-143通过促进HOXA10表达抑制子宫内膜异位发生的研究

摘　　要：目的构建MicroRNA-143慢病毒载体并探讨其在子宫内膜异位发生中的作用及机制。方法分离培养正位和异位子宫内膜基质细胞,检测HOXA10蛋白和MicroRNA-143的表达;构建MicroRNA-143慢病毒载体并感染子宫内膜基质细胞,流式检测HOXA10蛋白的表达。结果正位子宫内膜基质细胞HOXA10蛋白表达的平均荧光强度是异位子宫内膜基质细胞的1.74倍,差异有统计学意义(P<0.05);异位子宫内膜基质细胞内MicroRNA-143显著低于正位子宫内膜基质细胞,下降了64.56%,差异有统计学意义(P<0.05);MicroRNA-143转染可以显著促进异位子宫内膜基质细胞HOXA10蛋白表达,相对于阴性病毒组,平均荧光强度上升了72.03%,差异有统计学意义(P<0.05)。MicroRNA-143转染对正位子宫内膜基质细胞差异无统计学意义(P>0.05)。结论子宫内膜异位症患者MicroRNA-143和HOXA10蛋白表达显著下降,通过增强MicroRNA-143表达可以显著上调HOXA10蛋白表达抑制子宫内膜异位症的发生。Objective To construct MicroRNA-143 lentiviral vector, explore its role and the mechanism in occurrence of endometriosis. Methods Eutopic and ectopic endometrial stroma] cells were isolated and cultured, then the expressions of HOXA10 protein and MicroRNA-143 were detected. MicroRNA-143 lentiviral vector was constructed to transfect endometrial stromal cells. The expression of HOXA10 protein was detected by flow cytometry. Results The average fluorescence intensity of HOXA10 protein in eutopic endometrial stromal cells was 1.74 times more than that in ectopic endometrial stromal cells （P〈0. 05 ） . MicroRNA-143 level in ectopic endometrial stromal cells was statistically significantly lower than that in eutopic endometrial stromal cells, which decreased by 64. 56% （ P〈0. 05 ） . MicroRNA-143 transfection significantly promoted the expression of HOXA10 protein in ectopic endometrial stromal cells, compared with negative virus group, the average fluorescence intensity increased by 72. 03%, there was Statistically significant difference （P〈0. 05 ） . MicroRNA- 143 transfection had no significant impact on eutopic endometrial stromal cells （ P〉0. 05 ） . Conclusion The expressions of Mi- croRNA-143 and HOXA10 protein significantly decrease in the patients with endometriosis, enhancing MicroRNA-143 expression can inhibit the occurrence of endometriosis through upregulating the expression of HOXA10 protein.
