摘 要:目的:比较抗菌肽LL-37及其衍生物对肝癌细胞HepG-2增殖和凋亡的影响。方法:以肝癌细胞HepG-2为靶细胞,采用不同浓度抗菌肽LL-37及其衍生物处理后,通过四甲基偶氮噻唑蓝(MTT)比色法检测细胞的增殖情况,流式细胞术Annexin V/PI双染色测定细胞的凋亡情况。结果:随着抗菌肽LL-37及其衍生物作用浓度的增加和作用时间的延长,其对肝癌细胞增殖的抑制作用逐渐增强,二者的浓度都达到300μg/m L时作用72 h,对肝癌细胞增殖的抑制作用最大(P<0.01),且抗菌肽LL-37衍生物对肝癌细胞增殖的抑制率高于抗菌肽LL-37。流式检测结果显示:抗菌肽LL-37衍生物诱导的细胞凋亡率高于抗菌肽LL-37。结论:抗菌肽LL-37衍生物在抗肝癌细胞增殖和促进其凋亡方面的作用优于抗菌肽LL-37。Objective: To compare the effect of peptide LL-37 and its derivative on the proliferation and apoptosis of hepatocellular carcinoma HepG-2 cell line. Methods: The hepatocellular HepG-2 carcinoma cell line was used as targeted cells. MTT assay was applied to detect the effects of different concentrations of LL-37 peptide and its derivative on the proliferation of HepG-2 cells. And the flow cytometry annexin V/PI double staining was used to determine the cell apoptosis. Results: With the increase of the concentrations of LL-37 peptide and its derivative and the exposed time, the inhibitory rates on the proliferation of HepG-2 cells were gradually increased. The highest inhibitory rate occurred by LL-37 and its derivative with the concentration of 300 μg/mL after 72 h expose, and peptide LL-37 derivative induced a higher inhibitory rate on the proliferation of HepG-2 cells than that ofPeptide LL-37. The results of flow cytometry showed that the apoptosis rate induced by peptide LL-37 derivative was higher than that of peptide LL-37. Conclusion: Peptide LL-37 derivative could better enhance the apoptosis and inhibit the proliferation of HepG-2 cells than peptide LL-37.
