摘 要:目的探讨静电纺丝技术制备氧化锌/聚乙烯醇(zinc oxide/polyvinyl alcohol,ZnO/PVA)纳米纤维作为新型创伤敷料的可行性,考察其性能及对创伤愈合的促进作用。方法以溶液-凝胶法制备氧化锌纳米粒,再通过静电纺丝技术制备载有氧化锌纳米粒(1%、3%、5%)的ZnO/PVA纳米纤维并对其进行交联,采用扫描电镜和透射电镜对氧化锌纳米粒和纳米纤维进行观察,同时检测ZnO/PVA纳米纤维的透气性和吸水率,考察ZnO/PVA纳米纤维对金黄色葡萄球菌的抑制作用和对人皮肤成纤维细胞的毒性;建立大鼠皮肤创面模型,观察ZnO/PVA纳米纤维对创面的愈合作用。结果氧化锌纳米粒粒径为(64.69±20.47)nm,ZnO/PVA纳米纤维直径为(324.22±144.80)nm,纳米纤维直径较细且较为均一,交联效果明显;ZnO/PVA纳米纤维的透气性和吸水率均优于无纺布,差异具有统计学意义(P<0.05);ZnO/PVA纳米纤维对金葡萄的抑制作用较强;1%的ZnO/PVA纳米纤维无细胞毒性;ZnO/PVA纳米纤维能够促进创伤愈合,其促愈合作用优于创可贴(P<0.05)。结论采用静电纺丝技术制备的ZnO/PVA纳米纤维透气性好,吸水性强,抑菌作用较强,作用安全,能够促进创伤的愈合,有望成为理想的新型创伤敷料。Objective To prepare a new wound dressing based on zinc oxide/polyvinyl alcohol (ZnO/PVA) nanofibers by electrospinning, and determine its properties and effects on promoting wound healing. Methods The Zinc Oxide nanoparticles (ZnONPs) were prepared by a solution-gel method. ZnO/PVA nanofibers containing nano-Zinc Oxide (1%, 3% and 5% respectively) were prepared by electrospinning followed by a cross-linking process. The appearance features of ZnONPs and nanofibers were observed by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The permeability and water absorption of ZnO/PVA nanofibers were also measured. The antibacterial effect of ZnO/PVA nanofibers on Staphylococcus aureus and its cytotoxicity to human fibroblast cells were investigated. The skin wounds on rat model were established and the effect of ZnO/PVA nanofibers on the wound healing was investigated. Results The size of ZnONPs was 64.69±20.47 nm, and the diameter of ZnO/PVA nanofibers was 324.22±144.80 nm. The nanofibers were thin and uniform with obvious crosslinking, with better permeability and water absorption when compared with non-woven fabrics (P〈0.05). The results of bacteriostatic test showed that ZnO/PVA nanofibers had strong inhibitory effect on Staphylococcus aureus. No cytotoxicity was observed on human fibroblast cells by 1% ZnO/PVA nanofibers. In vivo study showed that ZnO/PVA nanofibers could promote wound healing and its promoting wound healing ability was better than woundplast. Conclusion Electrospinning-prepared ZnO/PVA nanofibers possess good permeability, strong water absorption and effective antimicrobial activity. ZnO/PVA nanofibers can promote wound healing, and are expected to be a new wound dressing.
