摘 要:目的:应用~1H-NMR法研究在知母醇提物干预下T2DM大鼠红细胞膜磷脂成分变化的相关性。方法:采用高糖高脂结合小剂量链脲佐菌素(STZ)腹腔注射诱导产生2型糖尿病模型。将造模成功大鼠随机分为模型组(DM组)、二甲双胍组(DMPG组)、知母醇提物高剂量组(H组)和知母醇提物低剂量组(L组),另设空白对照组。给药5周后,测量空腹血糖(FBG),并进行眼底后静脉丛取血,提取红细胞膜磷脂,采集各组红细胞膜磷脂的核磁共振氢谱。采用正交偏最小二乘法-判别分析(OPLS-DA)进行数据分析。结果:H组能明显降低糖尿病大鼠的空腹血糖(P<0.05)。OPLS-DA得分图显示DM组与NC组、DMPG组、H组的磷脂成分有明显差异。结论:在高剂量知母醇提物的干预下,二型糖尿病大鼠红细胞膜磷脂成分发生显著变化。Objective:To explore the relevance of different dosage of Rhizoma Anemarrhenae alcohol extracts intervened T2DM rats to the change of erythrocyte membrane phospholipid composition in T2DM rats by using 1H-NMR. Methods: Experimental rat model of Type 2 diabetes was built by the method of intraperitoneal injection of small dosesage (40mg/kg) of streptozotocin (STZ) and high fat diet inducing rat. The models were randomly divided into model group(DM group), metformin group (DMPG group) ; high-dosage of Rhizoma Anemarrhena alcohol extraction treated group (H group) ; low-dosage of alcohol extraction treated group(L group) ; and normal group(NC group). After five weeks administration, fasting blood glucose (FBG) was measured. Using retro- orbital venous plexus puncture to collect blood and phospholipids of erythrocyte membrane had being extracted. 1 H-NMR had being used to collect hydrogen spectrum of erythrocyte membrane phospholipids. Orthogonal Partial Least Squares Discriminate Analysis (OPLS-DA) for data analysis. Results: H group could significantly reduce fasting blood glucose in diabetic rats (P^0.05). OPLS- DA score plot display that phospholipid component of DM group shows significant difference between NC group, DMPG group and H group. Conclusion:After high-dosage of Rhizoma Anemarrhena alcohol extraction intervention, phospholipid composition of e- rythrocyte membrane display significantly difference in T2DM rats.
