摘 要:利用ISSR分子标记技术对广西特色蕉娄种质资源的遗传多样性进行分析。结果表明,从合成的35条ISSR引物中,筛选出13条条带清晰、多态性丰富及重复性好的引物,共扩增出96条条带,其中91条为多态性条带,平均多态性百分率达到93.69%。65份特色蕉类种质资源的遗传相似系数在0.479167~1000000之间,平均为0.804726。聚类分析结果表明,在遗传相似系数为0.71时,可以将基因型为ABB的大蕉及粉蕉与其他基因型的蕉类种质资源进行区分;遗传相似系数在0.74时,能够把大蕉和粉蕉区分开来。而在相似系数为0.76附近,大蕉种质资源聚成3类,在0.92附近时,粉蕉种质资源可聚为4类。Genetic diversity of 65 distinctive banana germplasm resources in Guangxi Zhuang Autonomous Region were studied by using ISSR markers in research. The results showed that 55 1SSR primers were screened, and 13 primers with clear band, more band number and good repeatability were selected. The 13 primers amplified 96 bands, out of which 91 bands were polymorphic, and the percentage of polymorphic loci was 93.69 %. Genetic similarity coefficient of 65 banana germplasm resources changed from 0. 479167 to 1. 000000, with an average of 0. 804726. Cluster analysis results indicated that ABB genotypes could be separated clearly from other genotypes at 0.71similarity level, and the plantain could be separated from dwarf bananas at 0.74. Then the plantain germplasm resources could be classified into three groups at 0.76 similarity level while dwarf bananas be four groups at 0.92.
