苏铁总黄酮对Lewis肺癌模型小鼠的抑制作用及免疫功能的影响

摘　　要：目的:探讨苏铁总黄酮对Lewis肺癌模型小鼠的抑制作用及免疫功能的影响。方法:选取造模成功的荷瘤小鼠84只,随机分为模型组、空白对照组、苏铁总黄酮高、中、低剂量组、顺铂组、联合组,每组12只。分别给药干预14d,于第15天摘眼球法取血,常规法剥取肿瘤组织、肺、脾、胸腺,并分别称重,计算抑瘤率、肺脏指数、脾脏指数和胸腺指数;采用ELISA法检测血清中IL-2、IL-10的含量。结果:苏铁高、中、低各剂量组、顺铂组和联合组小鼠瘤质量与模型组比较均降低,差异具有显著意义(P<0.01),苏铁总黄酮高、中、低各剂量组的脾脏指数和胸腺指数明显增加,且远远高于顺铂组与联合组。ELISA法检测结果显示,与模型组比较,苏铁总黄酮高中低各剂量组、顺铂组和联合组的IL-2水平明显升高,而IL-10水平除了苏铁总黄酮中剂量组之外的其他各组均明显升高。结论:苏铁总黄酮可以调节免疫细胞因子IL-2、IL-10的表达水平,抑制Lewis肺癌模型小鼠肿瘤细胞的生长与转移并提高其免疫功能。Objective： To explore the inhibition effect of total flavonoids from Cycas Revolute on the model mice with Lewis lung cancer,and its impacts on the immune function of tumor-bearing mice. Methods： 84 successful tumor-bearing mice were selected and then randomly divided into seven groups： model group,control group,high-dose group,medium-dose group,low-dose group,cisplatinum group,and combination group,12 mice in each group. Drug intervention for 14 days respectively,on the fifteenth day,pick the eyeball method for blood,use conventional methods to strip tumor tissues,lung,spleen and thymus and weight them,then calculate the inhibition rate,lung index,spleen index and thymus index. Using the enzyme-linked immunosorbent assay（ ELISA） to detect the IL-2 and IL-10 leves in serum. Results： Compared with the model group,the tumor weight of the mice from the high-dose group,medium-dose group,low-dose group,cisplatinum group,and combination group were decreased. The difference was highly significant（ P 0. 01）. The spleen index and thymus index of the high-dose group,medium-dose group and low-dose group were significantly increased,and far higher than the cisplatinum group and combination group. The ELISA results showed that compared with the model group,the IL-2 levels of the high-dose group,medium-dose group,low-dose group,cisplatinum group and combination group were significantly increased,and the IL-10 levels were significantly higher than the other groups in addition to the medium-dose group. Conclusion： The total flavonoids from Cycas Revolute may modulate the expression level of the IL-2 and IL-10,inhibit the growth and metastasis of the model mice with Lewis lung cancer and improve the immune function.
