摘 要:重组酶聚合酶扩增技术是一种可使微量核酸在体外高效快速扩增的新技术。与传统PCR及等温扩增技术相比较,其最显著的优势是在25℃-43℃条件下,5-20 min内观察到检测结果,且操作简单,设备成本低廉,被认为是一种可以代替PCR的新技术。本文综述了重组酶聚合酶扩增技术的原理和特点、检测方法及近十年来在国内外不同领域的应用情况,并展望了该技术在检测领域中的未来方向。Recombinase Polymerase Amplification(RPA) is a new kind of technique which can make micro-nucleic acid to be efficiently and rapidly amplified in vitro. Compared with the traditional techniques e.g.,polymerase chain reaction(PCR) and isothermal DNA amplification,RPA can easily observe the test results under the condition of 25 ℃-43 ℃ in 5 to 20 minutes. Due to its convenient operation and low cost,RPA has been considered as a new technique which can replace PCR. In this review,the principle,characteristic,detection method of RPA,and its application in different fields at home and abroad in the last decade are summarized. The application of RPA in inspection field is also prospected.
