摘 要:目的建立高效液相色谱分析法(HPLC)的方法测定瑞格列奈片中R(-)异构体的含量。方法使用AGP手性色谱柱(α-酸性糖蛋白键合硅胶作填料,4mm×100mm,5μm),以磷酸盐缓冲液:乙腈(85:15)为流动相,检测波长240nm,流速0.8mL/min。结果瑞格列奈与其R(-)异构体得到完全分离,在本方法所考察的浓度范围内相关性良好(R值〉0.99),回收率良好(95%~105%),检测限为0.2ng。结论HPLC法准确可靠、简便快速,可用于检测瑞格列奈中的R(-)异构体。Objective To establish the HPLC method to determinate the content of the R (-) isomer of repaglinide. Methods Using AGPChiral HPLC Column (Acid glycoprotein bonded silica gel as filler, 4 mm × 100 mm, 5 μm), phosphatebuffer:aeetonitrile (85 : 15 ) as mobile phase, detecting wavelength was 240 nm, flow rate was 0.8 mL/min. Results Repaglinide and itsR (-) isomer were separated completely, the relative factor was good (R 〉 0.99), the recovery rate was good (95% - 105%), the detecting limit was 0.2 ng. Conclusion This method is accurate, reliable, simple and fast, and it can be used to detect the R (-) isomer of repaglinide.
