摘  要:目的:建立不同产地加工方法浙贝母的高效液相色谱-电喷雾-质谱(HPLC-ESI-MS)的指纹图谱,并进行成分分析和鉴别。方法:液相采用Agilent Eclipse plus C18(4.6mm×250mm,5.0μm)色谱柱;流动相:溶剂A(10mmol/L甲酸铵溶液)和溶剂B(乙腈),梯度洗脱:0-45min,20%B;45-65min,20%-70%B;65-85min,70%-90%B;85-120min,90%B。柱温:30℃;流速:0.8m L/min;质谱采用电喷雾离子源,正离子全扫描检测;通过聚类分析和判别分析进行分类鉴别。结果:建立了不同产地加工方法浙贝母的HPLC-ESI-MS指纹图谱,分别得到38、31、31个共有峰,均推断出了17个化合物;聚类分析和判别分析均能很好地对不同产地加工方法的浙贝母进行分类。结论:所建立的HPLC-ESI-MS指纹图谱,为合理评价和鉴别浙贝母这3种不同产地加工方法提供了可靠手段,为全面控制浙贝母饮片的质量奠定基础。Objective: To establish fingerprints of thunberbg fritillary bulb from different producing areas by high performance liquid chromatography-electrospray ionization-mass spectrometry, and analyzed and identified the components. Methods: thunberbg fritillary bulb from different producing areas was performed on HPLC-ESI-MS using an Agilent Eclipse plus C18 column(4.6mm×250mm, 5.0 μm). The mobiles phase comprised solvent A(10mmol/L formic acid) and solvent B(acetonitrile). The gradient flow was as follows: 0-45 min, 20%B; 45-65 min, 20%-70%B; 65-85 min, 70%-90%B; 85-120 min, 90%B. The analysis was performed at a flow rate of 0.8 m L/min. The column temperature was 30℃. Mass spectrometry was tested by using electrospray ionization source and positive ion full scan. Classification and discrimination were performed by cluster analysis and discriminant analysis. Results: HPLC-ESI-MS fingerprints of thunberbg fritillary bulb were established in different producing areas, and 38, 31 and 31 common peaks were obtained, 17 compounds were deduced respectively. Clustering analysis and discriminant analysis can be very good for different processing methods of thunberbg fritillary bulb classification. Conclusion: The HPLC-ESI-MS fingerprints were established to provide a reliable method for the rational evaluation and identification of the three different processing methods of thunberbg fritillary bulb, and laid the foundation for the full control of quality thunberbg fritillary bulb pieces.
