摘 要:目的:研究健脾益智法对阿尔茨海默病(AD)大鼠海马区脑肠肽神经肽Y(NPY)m RNA表达水平的影响及其作用机制。方法:采用右侧海马注射凝聚态Aβ1-40的方法制作AD大鼠模型,以健脾益智混悬液、吡拉西坦片、0.9%氯化钠溶液分组灌胃给药连续治疗4周,采用半定量RT-PCR法检测比较各组大鼠海马区脑肠肽NPY m RNA的表达水平。结果:行为学评价分析得出,与模型对照组比较,中药组可以缩短Morris水迷宫实验逃避潜伏期,增加跨越平台次数。中药组AD大鼠海马区脑肠肽NPY m RNA表达水平明显高于模型对照组和西药组。结论:健脾益智法可以上调大鼠海马区脑肠肽NPY m RNA的表达水平,且上调作用明显优于西药组,对AD大鼠的学习记忆能力有明显的调节作用,为进一步研究及临床应用中医药防治AD提供实验依据。Objective: To study the effect of Jianpi Yizhi method on the expression of brain-gut peptide NPY m RNA in the hippocampus of rats with Alzheimer disease(AD) and to explore its mechanism. Methods: AD model rats were made by injecting condensed Aβ1-40 into the right hippocampus and rats in each group were given Jianpi Yizhi Decoction, Piracetam tablets and normal saline respectively by gavage for 4 weeks. Then the expression levels of brain-gut peptide NPY m RNA in the rats’ hippocampus were detected with semi-quantitative RT-PCR method. Results: As analyzed with behavioral evaluation, the Jianpi Yizhi method could shorten the rats’ escape latency in Morris water maze and increase the number of escaping. The expression level in the Jianpi Yizhi group was significantly higher than those in the model group and the Piracetam group. Conclusion: The Jianpi Yizhi method can up-regulate the expression of brain-gut peptide NPY m RNA in the hippocampus, with a significantly better effect than the Piracetam tablets. It also has a regulating effect on the learning and memorizing ability of rats. This paper can provide some experimental evidence for further studies on the clinical prevention and treatment of AD with traditional Chinese medicine.
