摘 要:目的:观察调和肝脾中药对模型大鼠气道反应性和相关细胞因子的影响。方法:将40只SD雄性大鼠随机平均分为4组:模型组、空白组、西医组、中药组。采用滴鼻法对实验大鼠进行MP(肺炎支原体)感染,后采用夹尾+冷冻+大黄灌胃多因素复合造成脾虚肝旺证模型;分别给药后,检测各组大鼠血清白细胞介素-4(IL-4)、干扰素γ(IFN-γ)含量;以不同浓度乙酰胆碱(Ach)经颈静脉注射刺激大鼠,测量吸气气道阻力(RL)、动态顺应性(Cdyn)及第0.1秒用力呼气容积(FEV0.1)等肺功能指标,反映动物气道反应性。结果:中药组、西医组、空白组分别与模型组比较,IL-4、INF-γ表达显著降低,中药组与西药组比,无显著差异;以较高浓度Ach(0.1、0.2mg/kg)激发大鼠时,中药组、西药组、空白组分别与模型组比较RL更低、Cdyn更高,但是中药组与西药组比较,差异无统计学意义;而FEV0.1则在各个时点,中药组、西药组、空白组较模型组均显著升高。结论:调和肝脾中药在改善肺功能和降低气道高反应性方面和西药作用相当,且具有越高质量浓度Ach刺激,改善作用越为明显的趋势。中药可以纠正INF-γ、IL-4免疫指标失衡,降低Th1及Th2类细胞因子的高表达,而且以降低Th2类因子更为明显。Objective: To observe the effects of Tiaohe Ganpi Formula on airway reactivity and related cytokines of model rats. Methods: Forty SD male rats were randomly divided into four groups: the model group, the control group, the Western medicine group and TCM group. The rats were infected with MP by the intranasal method, then the model of spleen weak and liver strong was established by tail squeezed + cryopreservation + rhubarb lavage. After drug administrated respectively, the contents of serum IL-4, IFN-γ in each group were detected, different concentration of acetylcholine(Ach) via jugular vein injection was used to stimulate rats, the animal airway reactivity including inspiratory airway resistance(RL), dynamic compliance(Cdyn) and forced expiratory volume in 0.1 seconds(FEV0.1) and other lung function indexes were measured. Results: Compared with the model group, the expression levels of IL-4 and INF-γ in the TCM group, western medicine group and control group were lower, and there was no statistical difference between TCM group and western medicine group. At a relatively high concentration of Ach(0.1, 0.2mg/kg) excitation in rats, compared with the model group, the RL in the TCM group, western medicine group and the control group was lower, and the Cdyn was higher, there were no statistical differences between TCM group and western medicine group. The scores of FEV0.1 in TCM group, western medicine group and control group at all time points were higher than model group. Conclusion: There was no differences between Tiaohe Ganpi Formula and the western medicine in improving lung function and reducing airway hyper-responsiveness, the higher the concentration stimulation of Ach, the better the improvement.
