骨髓间充质干细胞对器官移植中记忆性T细胞的影响

摘　　要：探讨骨髓间充质干细胞在器官移植中记忆性T淋巴细胞功能的影响。通过同种异基因皮肤移植的方法诱导CD8^+记忆性T淋巴细胞的产生,在体外应用混合淋巴增殖实验观察骨髓间充质干细胞对经过刺激后的T细胞增殖情况的影响;另一方面,通过同种异基因小鼠心脏移植模型的建立,在体内验证和观察骨髓间充质干细胞对小鼠移植器官生存寿命的影响。骨髓间充质干细胞在混合淋巴增殖实验中,可以有效抑制CD8^+记忆性T淋巴细胞的增殖能力,实验组淋巴细胞增殖指数明显低于对照组(t=4.575,p<0.05);在小鼠心脏移植模型中,输注骨髓间充质干细胞后移植心脏的生存寿命明显增加,差异有统计学意义(p<0.05)。骨髓间充质干细胞能够有效抑制器官移植中CD8^+记忆性T淋巴细胞的增殖,诱导免疫耐受,延长异体器官存活时间。To explore the effect of the mesenchymal stem cells on the memory T cells in the organ transplantation, CD8^＋ memory T cells from BalB/C mice were induced by allogeneic skin-transplantation, co-cultured with lymphocytes from C57 mice mixed by MSCs, observing the proliferation index ofT cells after being stimulated in vitro. On the other hand, the influence of the mesenchymal stem cells on the survival rates of transplanted hearts were proved in the mice heart-transplantation model. The proliferation of CD8^＋ memory T cells was significantly limited by the MSCs in the mixed lymphocyte proliferation test, and the proliferation index of experimental group was significantly lower than that in the control group （t=4.575, p〈0.05）. In the mice heart-transplantation model, the survival time of transplanted hearts was significantly lengthened after being injected with MSCs （p〈0.05）. So, MSCs can significantly decrease the proliferation of CD8^＋ mempry T cells, induce immune tolerance and lengthen the survival time of the transplanted organs.
