俞氏灌肠方对溃疡性结肠炎大鼠白介素-8、白介素-10和肿瘤坏死因子-α水平影响的研究

摘　　要：目的通过研究俞氏灌肠方对溃疡性结肠炎(UC)大鼠的影响,探讨其治疗作用及可能作用机制。方法将60只雄性大鼠随机分成正常组、模型组、SASP组、灌肠方低剂量组、中剂量组、高剂量组。除正常组外,其余组采用2,4,6-三硝基苯磺酸/酒精制备大鼠UC模型。ELISA法检测各组结肠黏膜组织的IL-8、IL-10、TNF-α表达水平,应用HE染色法检测肠黏膜组织病理学变化。结果与正常对照组比较,模型组中的IL-8、TNF-α含量明显上升,而IL-10明显降低,差异有统计学意义(P<0.05);与模型组比较,不同剂量治疗组中IL-8、TNF-α含量明显下降,差异均有统计学意义(P<0.05),而IL-10明显升高,并呈剂量相关性,差异有统计学意义(P<0.05)。结论俞氏灌肠方对溃疡性结肠炎大鼠可能通过降低促炎症因子IL-8、TNF-α的水平,升高抗炎因子IL-10的水平,修复肠黏膜而抑制溃疡性结肠炎的炎症反应。Objective To explore its therapeutic effect and the possible mechanism of Yu＇s Enema on ulcerative colitis（ UC）.Methods 60 male rats were randomly divided into the control group,model group,SASP group,and treatment groups with low,middle and high dose enema. Except for the control group,the other groups were used to establish the UC model with 2,4,6- trinitrobenzene sulfonic acid. The expression levels of IL- 8,IL- 10,TNF- α were tested by ELSIA,and the pathological changes were detected by HE staining in the colorectal mucosa. Results Compared with the normal group,the expression levels of IL- 8,TNF- α in the model group were higher than the control group,however,IL- 10 significantly reduced,and the difference was statistically significant（ P〈0. 05）; compared with the model group,IL- 8 and TNF- α expression levels in different dose treatment groups significantly reduced,with the differences statistically significant（ P〈0. 05）,while IL- 10 significantly increased,and also was dose- related,and the difference was statistically significant（ P〈0. 05）. Conclusion Yu＇s enema may repair the intestinal mucosa and inhibit the inflammatory response through reducing the levels of pro-inflammatory cytokines IL- 8,TNF- α,and elevating the level of anti- inflammatory cytokine IL- 10 to work on UC.
