回神颗粒含药脑脊液对Aβ_(25-35)所致PC12细胞损伤保护作用的研究

摘　　要：目的:观察回神颗粒含药脑脊液对Aβ_(25-35)所致PC12细胞损伤的保护作用。方法:采用PC12细胞建立24 h Aβ_(25-35)所致损伤模型。实验分成6组,即正常组,模型组,回神颗粒含药脑脊液高、中、低剂量组(20%、15%、5%),空白脑脊液组。将各剂量回神颗粒含药脑脊液与20μmol/L Aβ_(25-35)同时加入PC12细胞中培养24 h后,MTT法检测细胞活力。结果:回神颗粒含药脑脊液各剂量组均能显著提高Aβ_(25-35)所致受损PC12细胞的生存率,与模型组比较,差异均有显著意义(P<0.05)。结论:回神颗粒含药脑脊液能减轻Aβ_(25-35)所致的PC12细胞损伤,具有细胞保护作用。Objective： To evaluate the protective effects of Cerebrospinal Fluid containing Huishen Granule on injury of PC12 cells induced by Aβ_（25-35）. Methods： Alzheimer model of PC12 cells was induced by Aβ_（25-35） for 24 h. PC12 cells were divided into normal group,model group,blank cerebrospinal fluid（ 20%） group,and high,medium and low dosage cerebrospinal fluid containing Huishen Granule groups（ 20%,15%,5%）. Cerebrospinal fluid containing Huishen Granule groups were cultured with the corresponding concentrations of cerebrospinal fluid containing Huishen Granule and Aβ_（25-35） for 24 h.,then cell viability was determined by MTT assay. Results： Cerebrospinal fluid containing Huishen Granule at the concentrations of 20%,15%,5% significantly increased the cell viability of PC12 cells following Aβ_（25-35）injury（ vs model group,P〈0. 05）. Conclusion： Cerebrospinal fluid containing Huishen Granule could reduce the damage of PC12 cells induced by Aβ_（25-35）,and plays a protective role in the injured PC12 cells.
