摘 要:目的优化溃结灵结肠定位半浸膏片的提取工艺。方法采用单因素和星点设计-效应面法,乙醇浓度、乙醇倍量、提取时间为主要影响因素;以芍药苷、白术内酯Ⅰ和出膏率的综合评价为考察指标优化提取工艺。结果通过单因素法筛选出提取溶媒、提取方法、提取溶媒的浓度范围和提取次数;用星点设计-效应面法优选的最佳提取工艺为12倍量78%的乙醇提取2次,每次120 min。验证试验结果是为芍药苷的平均含量为29.39 mg·g-1、白术内酯Ⅰ平均含量1.396 mg·g-1、总得膏率为12.61%、总评OD为0.9446、预测OD值为0.949。结论本方法优选的提取工艺合理、可行、预测性良好。Objective To optimize the extraction of Kuijieling colon specific tablets. Methods Paeoniflorin, atractylenolide Ⅰand the paste rate of comprehensive evaluation were used as the index. The ethanol concentration, ethanol dosage and extraction time were used as the main influence factors. The extraction process of colon specific tablets was optimized by single factor and central composite design-response surface methodology. Results Optimum condition of extraction was extraction with12-fold, 78% ethanol twice 120 min each time. The average content of paeoniflorin was 29.39 mg·g-1, and the average content of atractylenolide Ⅰ was 1.396 mg·g-1; the total yield was 12.61%, general OD was 0.9446, and the predicted OD was 0.949. Conclusion The optimized extraction process is reasonable, feasible and predictive.
