摘 要:通过考察电泳分析条件对山莨菪中莨菪烷类生物碱分离和测定的影响,建立中药材山莨菪中阿托品、山莨菪碱、东莨菪碱和樟柳碱4种莨菪烷类生物碱非水毛细管电泳分析(NACE)的新方法.结果表明,在分离电压为28 k V、检测波长210 nm,缓冲体系为40 mmol·L^(-1)Tris溶液(水∶甲醇=1∶3,体积比,pH=7.0)的条件下,4种组分在7.5 min内实现基线分离,阿托品在5~70μg·m L^(-1)、樟柳碱在5~30μg·m L^(-1)、山莨菪碱和东莨菪碱在2~25μg·m L^(-1)的浓度范围内均具有良好的线性关系,检测限分别达到0.45、0.5、1.0、0.8μg·m L^(-1).该方法用于山莨菪中莨菪烷类生物碱的测定,结果令人满意.A capillary electrophoresis method has been developed for the determination of four tropane alkaloids,atropine,anisodamine,scopolamine,anisodine in Anisodus tanguticus after an investigation of basic factors which govern the nonaqueous capillary electrophoresis( NACE) separation of these alkaloids. The results showed that the four components could be well separated within 7. 5 min at a separation voltage of 28 k V in a 40 mmol·L^-1Tris system running buffer( water: methanol = 1 ∶ 3,volume ratio,pH = 7. 0) when the detection wave was set at 210 nm. A good linear relationship was observed in the range of 5 ~ 70,5 ~ 30 μg·m L^-1for atropine and anisodine respectively,2 ~ 25 μg·m L^-1for anisodamine and scopolamine. The detection limits were 0. 45,0. 5,1. 0,0. 8 μg·m L^-1for them. The method has been applied to the analysis of the four tropane alkaloids in Anisodus tanquticus,and the result was satisfactory.
