补肾化瘀方对宫腔粘连子宫内膜细胞生长周期及MMP-9的影响

摘　　要：目的:研究家兔补肾化瘀方含药血清对宫腔粘连子宫内膜基质细胞的生长周期及MMP-9因子的影响,探讨其部分作用机制。方法:选取宫腔粘连患者子宫内膜组织,制备宫腔粘连子宫内膜基质细胞模型。将家兔分为中药高、中、低剂量组及西药补佳乐组、空白组5组,用不同浓度补肾化瘀中药以及西药补佳乐灌胃7d,心脏采血并取含药血清,用家兔含药血清干预细胞模型,应用流式细胞检测仪检测细胞周期,ELISA法检测MMP-9因子的表达。结果:各组家兔含药血清作用于宫腔粘连患者子宫内膜基质细胞模型24h后,与空白组比较,各给药组的增殖指数(PI)和MMP-9含量均增高,凋亡指数(API)均降低,S期细胞百分比均增高,差异有统计学意义(P<0.01)。与西药组比较,中药各组PI增高,API降低,S期细胞百分比增高,中药中、低剂量组MMP-9表达减少,差异有统计学意义(P<0.01);但中药高剂量组MMP-9表达与西药组差异无统计学意义(P>0.05)。与中药中剂量组比较,中药高剂量组MMP-9表达增多,PI增高,API降低,S期细胞百分比增高,中药低剂量组PI降低,API增高,差异均有统计学意义(P<0.01);中药中、低剂量组的S期细胞百分比差异无统计学意义(P>0.05)。与中药低剂量组比较,中药高、中剂量组与西药组MMP-9表达增多,差异均具有统计学意义(P<0.01)。结论:含有补肾化瘀方的家兔含药血清可以提高宫腔粘连子宫内膜基质细胞中MMP-9因子的水平,促进宫腔粘连子宫内膜基质细胞的增殖,并降低其凋亡,不同浓度的补肾化瘀方对于宫腔粘连子宫内膜基质细胞的影响也有区别。Objective： To investigate the influence of serum containing Bushen Huayu prescription（ for tonifying the kidney and removing blood stasis） on the growth cycle of endometrial stroma cells and matrix metallopeptidase-9（ MMP-9） level in rabbits with intrauterine adhesion,as well as related mechanisms of action. Methods： The endometrial tissue was collected from patients with intrauterine adhesion to prepare a model of endometrial stroma cells. The rabbits were divided into high-,medium-,and low-dose traditional Chinese medicine（ TCM） groups,Western medicine Progynova group,and blank group. The rabbits were treated with various concentrations of Bushen Huayu prescription or Progynova by gavage for 7 days. Heart blood samples were collected,and the rabbit drug-containing serum was used for the intervention of the cell model. Flow cytometry was used to determine cell cycle,and ELISA was used to measure the expression of MMP-9. Results： After the model of endometrial stroma cells was treated by rabbit drug-containing serum for 24 hours,compared with the blank group,the treatment groups showed significant increases in proliferation index（ PI） and MMP-9 level,a significant reduction in apoptotic index（ API）,and a significant increase in the percentage of cells in S phase（ P 0. 01）. Compared with the Western medicine Progynova group,the TCM groups had an increase in PI,a reduction in API,and an increase in the percentage of cells in S phase,and the medium-and low-dose TCM groups had a significant reduction in the expression of MMP-9（ P 0. 01）. There were no significant differences in the expression of MMP-9 between the high-dose TCM group and the Western medicine Progynova group（ P 0. 05）. Compared with the medium-dose TCM group,the high-dose TCM group had significant increases in the expression of MMP-9 and PI,a significant reduction in API,and a significant increase in the percentage of cells in S phase,and the low-dose TCM group had a significant reduction in PI and a significant in
