摘  要:利用SRAP分子标记对来自四川省雅安、康定、泸定、西昌、成都和乐山6个地区的71份白三叶根瘤菌株的遗传多样性和遗传关系进行分析,结果表明:(1)23对引物组合共扩增出292条带,其中多态性条带279条,多态位点百分率95.55%;(2)单引物组合平均多态信息量(PIC)为0.341,条带信息指数(BI)为0.429,分辨力(RP)为6.226,标记指数(MI)为4.507;(3)供试菌株间遗传相似系数(GS)值介于0.493-0.962之间,平均为0.673;(4)采用UPGMA聚类方法,在GS=0.59时71份供试根瘤菌株可划分为3个组,地理来源相同或相似的菌株聚类结果相对集中;(5)方差分析(AMOVA)显示,在总的遗传变异中有85.05%发生在地理类群内,14.95%发生在地理类群间,均表现为极显著(P〈0.001)。表明SRAP标记对白三叶根瘤菌遗传变异的鉴别能力具有一定的可行性,是分析其遗传多样性的有效手段;供试白三叶根瘤菌株具有丰富的遗传多样性,且在分子水平上具有较远的亲缘关系;同时也揭示了不同地理环境根瘤菌株的遗传变异与其地域分布具有一定的相关性。Genetic diversity and relationship of 71 accessions of rhizobia were analyzed based on SRAP markers.These rhizobia were isolated from wild white clovers which were collected from six different sampling sites(Ya'an,Kangding,Luding,Xichang,Chengdu and Leshan)in Sichuan province in China.The results showed that:(1)A total of 292 bands were detected by using 23 primer pairs,and 279bands(95.55%)were polymorphic loci.(2)The average percentage of polymorphism information content(PIC),band informativeness(BI),resolving power(RP),marker index(MI)was 0.341,0.429,6.226 and 4.507,respectively.(3)Genetic similarity(GS)among accessions ranged from 0.493 to 0.962 with an average of 0.673.(4)Based on the UPGMA cluster analysis,71 accessions were divided into three groups at GS=0.59,and the accessions collected from the same or similar geographical origin clustered together to a great extent.(5)85.05% genetic variation was observed among geographical groups,and 14.95% among geographical groups according to the molecular variance analysis(AMOVA).These results suggested that SRAP maker had a good ability to identify the rhizobia of white clover and was an effective way for analyzing genetic diversity of rhizobia.The tested rhizobia had rich genetic diversity and smaller phylogenetic relationship at molecular level.Moreover,the results also revealed that the genetic variation of Rhizobium strains in different geographical groups was associated with their geographical distributions.
