摘 要:目的:确认苦参中总黄酮最佳超声提取工艺及其对多种病原菌的最小抑菌浓度(MIC)。方法:采用L18(37)正交实验设计方法,确定超声提取优化工艺,以微量稀释法考察苦参总黄酮水溶液的MIC。结果:最佳超声提取条件为1∶10料液比,50%乙醇浓度,提取15 min,p H=9,提取1次,MIC结果表明苦参总黄酮对多种病原菌均有一定抑制效果。结论:超声提取法具有快速、操作简单、节省能源等优点,此工艺可用于实验室提取苦参总黄酮实验,其对金黄色葡萄球菌、大肠埃希菌、变形杆菌、肺炎克雷伯菌均有一定抑制效果,对铜绿假单胞菌效果不明显。Objective:To explore the optimized ultrasonic extraction condition for total flavonoids in Radix Sophorae Flavescentis and the minimal inhibitory concentration(MIC) for various pathogenic bacteria. Meth ods:L18(37) orthogonal experimental method was used to determine the optimized ultrasonic extraction technology,and MIC was determined through micro-dilution method. Results:The optimized ultrasonic extraction conditions were as follows:ratio of material to solution was 1 ∶10;50% ethanol concentration,15 minutes for extraction,p H value 9,and extracted for 1 time. MIC test showed that the total flavonoids from Radix Sophorae Flavescentis had certain inhibitory effect on various pathogenic bacteria. Conclusion:Ultrasonic extraction method was fast,easy to operate,and energy saving. This process can be used to extract the total flavonoids from Radix Sophorae Flavescentis in laboratory. Total flavonoids from Radix Sophorae Flavescentis showed certain inhibitory effect on Staphylococcus aureus,Escherichia coli,Bacillus subtilis,Klebsiella pneumonia,but the effect on Pseudomonas aeruginosa was not obvious.
