摘 要:目的探究高脂血症小鼠海马CA3区神经元形态、尼氏体含量以及CaMK Ⅱ蛋白表达的情况。方法20只ApoE-/-鼠随机分为对照组(n=10)和高脂血症组(n=10),分别饲养普通饲料和高脂饲料。分别采用HE染色和尼氏染色观察海马CA3区形态学变化,免疫组织化学方法观察各组小鼠海马神经元内CaMK Ⅱ蛋白表达情况。结果 HE染色结果显示,高脂组和对照组海马神经元形态无明显差异;尼氏染色结果显示,高脂组尼氏体含量较正常组减少;免疫组化结果表明,高脂组海马CA3区细胞数目减少,染色较深,CaMK Ⅱ阳性蛋白的表达较正常组明显减少。结论高脂血症可导致小鼠海马CA3区神经元CaMK Ⅱ蛋白表达降低。Objective To investigate morphology of neurons and Nissl body content and CaMK Ⅱ expression in hippoeampal CA3 region of mice with hyperlipidemia. Methods Twenty ApoE-/- mice were randomly divided into hyperlipidemia group(given high fat diet) and control group(normal diet). HE staining and Nissl staining were used to observe hippocampal neuron morphology, CaMK Ⅱ expression was measured by immunohistochemistry. Results There are not obviously difference in hippocampal neuron morphology among two groups. Nissl body content in hippocampus CA3 region was significantly reduced when mice were fed with hyperlipidemia. Cell number was obviously reduced and cells were hyperchromatic in CA3 region after hyperlipidemia. In control group, a strong CaMK Ⅱ immunoreaetivity was observed in CA3 region. Of interest, CaMK Ⅱ positive cell was obviously decreased in CA3 region after hyperlipidemia. Conclusion Hyperlipidemia can decrease the expression of CaMK Ⅱ protein in the neurons of CA3 region.
