祛风活血丸对EAU大鼠模型Fas／FasL表达的影响

摘　　要：目的探讨祛风活血丸对实验性自身免疫性葡萄膜炎(EAU)大鼠血清Fas、FasL水平的影响。方法将70只Lewis大鼠随机分为空白组(A组),模型对照组(B组),阳性对照组(C组),以及祛风活血丸低、中、高剂量组(D、E、F组)。采用大鼠右后足垫注射含IRBP(光感受器间维生素A类结合蛋白)的PBS溶液+完全弗氏佐剂制作EAU模型,A组大鼠注射无菌生理盐水。造模后各组大鼠灌胃给药,A、B组予生理盐水,C组予熊胆开明片,D、E、F组分别予低、中、高剂量的祛风活血丸。观察大鼠眼部情况变化,并采用酶联免疫吸附试验(ELISA)法检测给药后第7天、14天、21天、28天时各组大鼠的Fas、FasL血清浓度。结果 1.成功诱发Lewis大鼠的EAU模型,在免疫后不同时间发生临床可见的炎症,造模成功率91.7%(55/60),空白组大鼠无临床可见葡萄膜炎表现。2.空白组各时间点的Fas、FasL血清浓度始终位于较低水平,模型对照组各时间点的Fas、FasL水平均为最高,明显高于空白组(P<0.01),并有随时间延长逐渐下降的趋势。中、高剂量组各时间点的Fas、FasL水平较为接近,低于同期低剂量组及阳性对照组,与模型组比较下降更为明显(P<0.05)。结论Fas/FasL介导的细胞凋亡对EAU发病具有重要意义,祛风活血丸能使大鼠血清中Fas、Fas L含量明显降低。OBJECTIVE Explore the functions of huoxue pill on experimental autoimmune uveitis （EAU） serum level of Fas and FasL in rats. METHODS 0 Lewis rats were randomly divided into blank group （group A）, model control group （group B）, positive control group （group C）, as well as the functions of huoxue pill low, medium and high dose group （group D, E, F）. Using the right rear foot pad injection in rats including IRBP （vitamin A bind- ing protein） between photoreceptor of PBS solution ＋ make EAU model complete freundls adjuvant,group A sterile saline injection in rats. After building groups of rats lavage, A, B group with physiological saline, group C to bear bile enlightened, D, E, F group to low, medium and high doses of the functions of huoxue pellet. Observation rat eye situ- ation changes, and using enzyme-linked immunosorbent assay （ELISA） method to detect drug delivery after 7 days, 14 days ,21 days ,28 days between groups of Fas and FasL in rats serum concentration. RESULTS 1. The EAU suc- cessfully induce Lewis rats model, clinical visible at different time before and after the immune inflammation, build- ing success rate 91.7% （55/60） ,blank group rats without clinical visible uveitis. 2. Each time point blank group serum concentrations of Fas and FasL always located in the lower level, model control group of each time point was the highest level of Fas and FasL, significantly higher than the blank group （P〈0.01）, and has a tendency to extend gradually decreases with time. Each point in time, the high dose group of Fas and FasL level is relatively close, lower than the same period and low dose group and positive control group, compared with model group decreased more significantly （P〈0.05）. CONCLUSIONS Fas/FasL mediated apoptosis of EAU incidence is of great significance and functions of huoxue pill can make the rats serum content of Fas and FasL decreased obviously.
