摘 要:目的:探讨褪黑素(MT)对脂多糖(LPS)诱导的脓毒症大鼠肺损伤组织的保护作用。方法:成年清洁级SD雄性大鼠72只,随机分为对照组(静脉注射0.9%氯化钠溶液)、LPS组(静脉注射LPS 1 mg/kg)、LPS+MT1组(静脉注射MT0.1mg/kg+LPS 1mg/kg)、LPS+MT2组(静脉注射MT 1mg/kg+LPS 1mg/kg),每组18只。给药后6h取血,应用酶联免疫吸附法(ELISA)检测各组大鼠血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)和IL-10的含量;取大鼠肺组织,匀浆,测定超氧化物歧化酶(SOD)、丙二醛(MDA)和髓过氧化物酶(MPO)含量。结果:LPS组大鼠血清TNF-α、IL-6和IL-10水平较对照组显著升高(P<0.01);给予MT干预后,大鼠TNF-α、IL-6水平下降而IL-10水平上升(P<0.05),且这一效应随MT剂量增加而增大。LPS组大鼠肺组织SOD含量较对照组明降低,而MDA和MPO含量显著升高(P<0.01);给予MT干预后,大鼠SOD水平有显著升高,MDA和MPO水平明显下降(P<0.05),且这一效应随MT剂量增加而增大。结论:MT可降低LPS诱导的脓毒症大鼠的炎症反应,并能有效拮抗脂质过氧化造成的肺组织损伤,起到一定的肺保护作用;MT对肺损伤组织的保护作用与其剂量正相关。Objective: To investigate the protective effects of melatonin(MT)on lung injury induced by lipopolysaccharide(LPS)in rats with sepsis.Methods:Seventy-two rats were randomly divided into four groups with 18 rats in each group:control group(0.9% NS,i.v.),LPS group(LPS 1mg/kg,i.v.),LPS+MT1group(MT 0.1mg/kg+LPS1mg/kg,i.v.)and LPS+MT2group(MT 1mg/kg+LPS 1mg/kg,i.v.).Carotid arterial blood of all rats was collected at6 hafter administration and serum levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-10(IL-10)were detected by enzyme linked immunosorbent assay(ELISA).The lung homogenate contents of superoxide dismutase(SOD),malondialdehyde(MDA)and myeloperoxidase(MPO)were also examined.Results:Compared with control group,the levels of serum TNF-α,IL-6and IL-10 were significant higher in LPS group(P〈0.01).With the MT intervention,the levels of TNF-αand IL-6were significantly decreased but the IL-10 level was remarkably increased(P〈0.05).This effect was more remarkable with the increase of the dose of MT.The level of SOD in LPS group was remarkably lower than in control group,but the levels of MDA and MPO were significantly higher(P〈0.01).Compared with LPS group,the level of SOD in LPS+MT1group was significantly increased,however,the contents of MDA and MPO were decreased(P〈0.05).The same trend was more prominent in LPS+MT2group.Conclusions:MT can decrease the lung injury by reducing the inflammatory response and lipid peroxidation induced by LPS in rats with sepsis.The protective effect was positively associated with the dose of MT.
