摘  要:目的探讨多位点序列分析技术在腹泻病原空肠弯曲菌分子溯源方面的运用。方法用多位点序列分析(MLST)对上海市浦东新区腹泻病原监测中的70株阳性空肠弯曲菌菌株的7个管家基因进行扩增、测序并上传至MLST数据库确定基因序列型(ST型),并运用Bio Numerics 7.1构建遗传进化树和聚类图进行亲缘性分析。结果 70株空肠弯曲菌分为52种已知ST型,分属13个同源谱系,ST353同源复合体占了流行株的26%。从70株分型结果中能找到聚集性病例。结论运用MLST技术能在分子水平上对空肠弯曲菌进行分型,并能直观表现菌株间的相互关系,从散发病例中找到聚集特征,为疾病的控制提供依据。[Objective]To explore the application of Multilocus sequence typing(MLST) in molecular traceability of Campylobacter. jejuni.[Methods]70 strains of Campylobacter. Jejuni were isolated from diarrhea stool specimens in Pudong New Area of shanghai,and 7 house keeping genes were amplified and sequenced. The nucleotide sequences were submitted to the database to obtain the allele numbers and sequence types(STs). Finally,Bio Numerics 7.1 was used to construct the phylogenetic trees and cluster plots.[Results]70 strains were classified into 52 STs which belonged to 13 Clonal Complexs. The most prevalent sequence type(ST) was ST353(26.0%). The clustering cases had been found.[Conclusion]The relationships between species of Campylobacter. Jejuni at the level of genes are accurately determined by MLST,the pattern variation provides a useful method for linking a particular strain to its source.
