摘 要:为探究不同抗氧化剂对鸡胚盘细胞(blastodermal cells,BCs)冷冻保存效果,试验在冷冻液中分别添加不同浓度的N-乙酰半胱氨酸(N-acetyl-L-cysteine,NAC)、过氧化氢酶(catalase,CAT)和番茄红素(lycopene,LP)3种抗氧化剂,用胚胎程序冷冻仪冻存BCs,解冻后测定细胞活率(简称"活率"),并于培养24h后测定细胞贴壁率(简称"贴壁率")。结果表明:在活率上,除了0.05mol/L NAC组(76.7%)与800U/mL CAT组(77.3%)极显著低于对照组外(P<0.01),其余各组与对照组差异均不显著(P>0.05);在贴壁率上,除了800U/mL CAT组(34.2%)极显著低于对照组(P<0.01)外,其余试验组均极显著高于对照组(P<0.01),其中NAC、CAT与LP的最佳浓度,分别为0.01mol/L(48.3%)、200U/mL(45.4%)与0.05g/L(39.8%)。因此,在冷冻液中添加适量的NAC、CAT或LP,可以提高BCs冷冻保存活率(差异不显著),极显著提高解冻BCs培养24h后的贴壁率。The different concentration of N-acetyl-L-cysteine (NAC),catalase (CAT) and lycopene (LP) were added into the cryoprotectant respectively,in order to investigating the effect of different antioxidants on cryopreservation of the chicken blastodermal cells (BCs). The BCs were frozen by the program freezer, together with the different kinds and concentrations of cryoprotectants. The cells viability (CV) and the cell adherent rate cultured for 24 h (CAR) were determined after thawing. The results showed that for the CV,only group NAC with 0. 05 mol/L(76. 7%)and group CAT with 800 U/mL(77.3%) were lower than control group and had extremely significant difference (P〈0.01). The rest groups, such as group NAC, CAT and LP all had not significant difference with control group (P〉0.05). For the CAR,the group CAT with 800 U/mL was lower than the group control (34.2%), the rest groups were all higher than control group and had extremely significant difference (P 〈0. 01). The best concentration of NAC was 0. 01 mol/L (48.3%) ,CAT was 200 U/mL (45.4%),LP was 0.05 g/L (39.8%). Therefore,the addition of NAC,CAT or LP into the cryoprotectant for freezing BCs, could not only improve the CV (not significant difference),but also the CAR (extremely significant difference).
