摘  要:目的建立一种快速、准确的龟甲及其伪品随机扩增DNA多态性(RAPD)鉴定方法,为龟甲的鉴别提供可靠依据.方法采用盐析法从龟甲及其伪品中提取mt DNA,并进行随机DNA多态性扩增.结果通过电泳图谱明显看出正品龟甲与其常见伪品mt DNA随机扩增产物条带数量和位置均不同,说明本文建立的方法可以有效区分正品龟甲与其常见伪品.结论 RAPD技术为龟甲及其伪品的鉴别提供一种简单、有效、可信度高的方法.Objective To establish a rapid and accurate method for the identification of random amplified polymorphic DNA (RAPD) of tortoise shell and its counterfeits, and provide a reliable basis for the identification of tortoise shell. Method The mt DNA from tortoise shell and its counterfeits was separated by salting out method and its random DNA polymorphism amplification was carried out. Results The electrophoretogram clearly showed the differences in the number and location of rat DNA random amplified bands between genuine tortoiseshell and its counterfeits, indicating that this method can effectively distinguish the genuine tortoise shell and its counterfeits. Conclusion RAPD technology can provide a simple, effective and highly reliable method for the identification of tortoise shell and its counterfeits.
