摘 要:目的研究线粒体磷酸转载体(Pi C)在大鼠缺血性心肌损伤中的表达情况及姜黄素的保护作用。方法36只SD大鼠,随机分成3组。对照组:接受生理盐水作为干预因素;心肌损伤组:垂体后叶素25 U/kg尾静脉注射,每日一次,4周后改成垂体后叶素25 U/kg尾静脉注射,隔日一次,共4周;姜黄素组:在每次垂体后叶素半小时内应用姜黄素,以30 mg/kg灌胃。第11周时将所有大鼠处死,解剖取心脏组织进行心脏指数(HVW/BW)、细胞凋亡和Western blot检测。结果心肌损伤组大鼠体重均低于对照组,但HVW/BW显著高于对照组,差异有显著性(P<0.05);凋亡检测发现心肌损伤组心肌细胞凋亡率明显高于对照组及姜黄素干预组(P<0.05);心肌损伤组的Pi C蛋白表达情况明显高于对照组及姜黄素干预组(P<0.05),姜黄素进行干预后,Pi C表达情况明显下降,但仍高于对照组,差异均具有统计学意义(P<0.05)。结论 Pi C在缺血性大鼠心肌损伤中的表达上调,姜黄素对心肌细胞损伤具有保护作用,下调Pi C的表达。Objective To study the expression of phosphate carrier( Pi C) in ischemic myocardial injury in rats and the protective effect of curcumin. Methods 36 SD rats were randomly divided into 3 groups,control group received saline as intervention factor; myocardial injury group: posterior leaf( 25 U / kg) intravenous injection,once a day,for four weeks,later changed into a pituitary leaf( 25 U / kg) intravenous injection,once every other day,a total of around; curcumin group: curcumin was applied to the 30 mg / kg of the stomach in a half hour after each of the pituitary,all rats were sacrificed at eleventh weeks,anatomy of heart tissue to HVW / BW ratio,cell apoptosis and Western blot. Results The body weight of rats in myocardial injury group was lower than that of the control group,but the HVW / BW ratio was significantly higher than that in the control group( P〈0. 05). The apoptosis rate of myocardial cells in myocardial injury group was significantly higher than that in the control group( P〈0. 05). The expression of Pi C in myocardial injury group was significantly higher than that in the control group and the curcumin intervention group( P〈0. 05). When given curcumin intervention,Pi C expression was significantly decreased,but still higher than the control group,the difference was statistically significant( P〈0. 05). Conclusion The expression of Pi C in ischemic myocardium of rats with myocardial injury was up- regulated,and the protective effect of curcumin on myocardial cell injury was decreased,and the expression of Pi C was down regulated.
