摘 要:目的研究降脂宁提取物对血管钙化的影响及其可能的机制。方法将三月龄雄性Wistar大鼠40只依据随机数字表法分为对照组、高脂钙化组、降脂宁组、阿托伐他汀组和复药组(降脂宁+阿托伐他汀),每组8只。15周后取血检测血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)和低密度脂蛋白(LDL);取胸和腹主动脉分别测定血管钙、丙二醛(MDA)、一氧化氮(NO)含量及碱性磷酸酶(ALP)和超氧化物歧化酶(SOD)活性;同时,各组大鼠取胸主动脉用TUNEL法测定细胞凋亡率,用Western blot检测caspase-3蛋白表达水平。结果与对照组比较,高脂钙化组血清TG和TC含量均明显升高(P<0.01);阿托伐他汀组及复药组血清TG、TC和LDL,降脂宁组血清TC均明显低于高脂钙化组(P<0.01);复药组血清LDL明显低于对照组、高脂钙化组和降脂宁组(P<0.01)。与对照组比较,高脂钙化组血管钙含量和ALP活性明显升高(P<0.05或P<0.01);与高脂钙化组比较,降脂宁组血管钙含量和ALP活性明显降低(P<0.05或P<0.01),复药组血管ALP活性明显降低(P<0.01)。与对照组比较,高脂钙化组血管MDA含量明显升高(P<0.01),而复药组明显低于其他四组(P<0.01);与对照组比较,高脂钙化组血管NO含量明显降低(P<0.01)。与对照组比较,高脂钙化组血管平滑肌细胞凋亡率明显升高(P<0.01);各给药组血管平滑肌细胞凋亡率均明显低于高脂钙化组(P<0.01)。与对照组比较,高脂钙化组血管caspase-3灰度比明显升高(P<0.01);各给药组血管caspase-3灰度比明显低于高脂钙化组(P<0.01)。结论降脂宁提取物和阿托伐他汀均有明确的降脂疗效,而两药结合降低LDL的效果尤为明显,其作用可能与抗氧化作用有关;降脂宁组和复药组均能降低血管钙含量和ALP活性,具有一定的抑制血管钙化作用,其作用可能与其抑制血管平滑肌细胞凋亡和下调caspase-3的表达水平有关。Objective To study the effects and mechanism of Jiangzhining extract on vascular calcification. Methods Three months old male Wistar rats (n=40) were divided into control group, high-fat calcification group, Jiangzhining group, Atorvastatin group and mixture group (Jiangzhining plus Atorvastatin) by using random number table method, each group was 8 rats. After 15 weeks, the levels of serum total cholesterol (TC), triglyceride (TG), high-density lipoprotein (HDL) and low density lipoprotein (LDL) were detected, the contents of vascular calcium, malondialdehyde (MDA) and nitric oxide (NO) were observed and activity of alkaline phosphatase (ALP) and superoxide dismutase (SOD) were measured on the chest and abdomen aorta. The apoptosis rates of the vascular smoothmuscle cells were observed via in situ apop- tosis detection (TUNEL) and the protein expression level of caspase-3 was detected by Western blot. Results Compared with control group, the levels of TG and TC in high-fat calcification group were significantly increased (P 〈 0.01). The levels of serum TG, TC and LDL in Atorvastatin group and mixture group were significantly lower than those in high-fat calcification group, level of serum TC in Jiangzhining group was significantly lower than that in high-fat calcification group (P 〈 0.01). The level of serum LDL in mixture group was significantly lower than those in control group, high-fat calcification group and Jiangzhining group (P 〈 0.01). Compared with control group, vascular calcium content and ALP activity in high-fat calcification group were increased significantly (P 〈 0.05 or P 〈 0.01). Compared with high-fat calcification group, vascular calcium content and ALP activity in Jiangzhining group were decreased significantly (P 〈 0.05 or P 〈 0.01), ALP activity in mixture group was decreased significantly (P 〈 0.01).
