摘 要:目的研究脾酪氨酸激酶(spleen tyrosine kinase,Sy K)和肝细胞生长因子(hepatocyte growth factor,HGF)在乳腺癌表达及临床意义。方法采用免疫组织化学方法分别检测120例乳腺癌组织、100例正常乳腺组织、100例纤维腺瘤组织之间的Sy K和HGF蛋白表达,分析乳腺癌组织中二者的表达水平与临床病理学特征的关系。结果 120例乳腺癌组织中Sy K、HGF的表达率分别为30.0%、68.3%,100例正常乳腺组织表达率分别为60%、33%,100例纤维腺瘤组织表达率分别为63%、36%,Sy K、HGF的表达率在乳腺癌组织与其他两组之间比较,差异有统计学意义(P<0.01);而二者表达率在正常乳腺组织与纤维腺瘤组织之间比较,差异无统计学意义(P>0.05)。Sy K、HGF的表达与乳腺癌临床分期、淋巴结转移相关(P<0.05),与年龄、肿块直径及脉管侵犯无关(P>0.05)。随乳腺癌临床分期越晚、有淋巴结转移者,Sy K表达率缺失越明显,HGF的表达水平依次升高,差异有统计学意义(P<0.05)。在乳腺癌中Sy K与HGF之间表达呈负相关(r=-0.649,P<0.01)。结论 Sy K的表达缺失及HGF在乳腺癌的表达增高与乳腺癌的增殖、侵袭和转移等生物学行为密切相关,联合检测Sy K、HGF在乳腺癌组织中的表达情况有助于乳腺癌的早期诊断及预后的判断。Objective To study expression and Clinical Significance of SyK and HGF in breast cancer. Methods The expressions of SyK and HGF protein were detected by immunohistochemical method in 120 samples of breast cancer, 100 samples of normal breast tis- sue and 100 samples of fibroadenoma,analyzed its relationship with assays showed that the positive expression rate of SyK and HGF were 30.0% , 68.3% respectively in 120 breast cancer, whereas in normal breast tissues of 100 cases the positive expression rate were 60% , 33% and in fibroadenoma tissues of 100 cases the positive expression rate were 63% , 36%. There were statistically significant differences between the expression levels of Syk and HGF of breast cancer, normal breast tissue and fibroadenoma ( P 〈 O. O1 ). There were no statisti- cally significant differences between the expression levels of Syk and HGF of normal breast tissue and fibroadenoma ( P 〉 0. 05 ). These da- ta showed that the high expression of SyK and HGF is remarkably related to the clinincal stage and lymph node metastasis( P 〈 O. 05), but not to the age and tumor size and vascular invasion. In breast cancer, the SyK no expressions were associated with advanced clinincal stage and lymph node metastasis. There were statistically significant differences in the positive expression of HGF increased remarkably from breast cancer( P 〈 0.05 ). The expression of SyK and HGF were negative correlated ( r = - O. 649,P 〈 0.01 ). Conclusion We con- cluded that both the deficiency of SyK and the high expression of HGF had a close relevance to the malignant biological behaviors including proliferation, penetration and metastasis of breast cancer. The exploration for SyK and HGF could be useful for evaluating the early diagnosis and prognosis of breast cancer.
