摘 要:目的:观察夹脊电针对急性脊髓损伤大鼠脊髓组织神经干细胞增殖分化的影响,探究其与急性脊髓损伤后神经细胞再生的关系。方法:选用72只体重在200-220 g之间的雄性SD大鼠。随机将大鼠分为假手术组,模型组,电针组和药物组,每组分术后1 d、7 d、14 d等时间点,每个时间点有6只。采用改良的Allen法建立大鼠T9-T10段脊髓急性中度损伤模型,用免疫荧光双标法检测各组大鼠脊髓组织中Brd U/nestin阳性细胞的共表达,用BBB评分反映大鼠的运动功能恢复。结果:治疗结束后,模型组、电针组和药物组BBB评分和Brd U/nestin阳性细胞数呈逐级增高趋势,其中与模型组相比,电针组Brd U/nestin阳性细胞数明显增多(P〈0.05),BBB评分明显提高(P〈0.05)。结论:夹脊电针干预后,Brd U/nestin表达增多,BBB评分提高,促进了大鼠神经干细胞的分化和运动功能的恢复。Objective:To observe the effect of Jiaji electro-acupuncture on acute spinal cord injury in rats neural stem cell proliferation and differentiation and explore its relationship with the nerve cell regeneration after acute spinal cord injury.Method: Seventy-two male SD rats weighing between 200 - 220 g were randomly divided into sham operation group,model group,electro-acupuncture group and drug group,each group was divided into 3 time points 1,7 and 14 days after operatin,each time points have 6 rats.Spinal cord injury model was made at T9-T10 segments with Modified Allen method.Double immunofluorescence detection was used to detect Brd U/nestin co-expression in spinal cord injury tissue and the BBB scale after spinal cord injury to assess the motor function.Result: At the end of treatment,model group,electro-acupuncture group and drug group's BBB rating and Brd U/nestin-positive cells had a progressively increasing trend.Compared with the model group,EA group's Brd U/nestin-positive cells were significantly increased(P〈0.05) and BBB score significantly improved(P〈0.05).Conclusion: After Jiaji electro-acupuncture intervention,Brd U/nestin expression was increased,BBB ratings were improved and the differentiation of neural stem cells and motor function were promoted.
