摘  要:目的:建立不同等级延胡索药材HPLC指纹图谱,并同时测定原阿片碱、盐酸巴马汀、盐酸小檗碱、去氢紫堇碱、延胡索乙素和紫堇碱6种主要成分的含量,对不同等级延胡索药材质量进行研究。方法:对延胡索药材等级分类,采用Agilent Eclipse XDB-C18(4.6μm×250 mm,5μm),乙腈-0.6%醋酸溶液(p H5.0)为流动相进行梯度洗脱,检测波长280 nm。对10批3个等级延胡索药材进行测定,运用国家药典委员会"中药色谱指纹图谱相似度评价软件"2004A版进行评价,并对6种成分进行含量测定。结果:含量测定条件通过方法学验证,精密度、线性关系、准确度、稳定性、重现性良好;建立了延胡索生物碱类成分HPLC指纹图谱。结论:该法所建立的延胡索生物碱类成分指纹图谱特征性强、方法简便,结合6个生物碱含量测定可更好地控制其质量,对延胡索等级分类具有指导意义和参考价值。Objective: To establish a method for HPLC fingerprint determination of different grade of Rhizoma corydalis and simultaneous determine the contents of Protopine,Palmatine hydrochloride,Berberine hydrochloride,Dehydrocorydaline,Tetrahydropalmatine and Corydaline in Rhizoma corydalis and further study the quality standard of different grades of Rhizoma corydalis.Methods: With the different grades of Rhizoma corydalis,the optimal conditions of separation and detection were achieved on a Agilent Eclipse XDB-C18(4.6 μm × 250 mm,5 μm) with linear gradient elution of acetonitrile-0.6% acetate solution(p H = 5.0).The detection wavelength was 280 nm.Collect 10 batches 3 grades of Rhizoma corydalis to assess the similarities according to similarity for Chromatographic Fingerprint of Traditional Chinese Medicine(2004A).Results: Chromatography condition on determining the alkaloids contents in Rhizoma corydalis by HPLC was advantageous.The HPLC fingerprint of different grades of Rhizoma corydalis was established.Conclusion: This study indicated that the combination of quantitative and chromatographic fingerprint analysis can be readily utilized as the quality control method for different graded of Rhizoma corydalis.
