人参皂苷干预酒精性脂肪肝形成的实验研究及对小鼠抗氧化应激的影响  被引量:7

摘  要:目的:观察人参皂苷对酒精性脂肪肝(AFL)小鼠肝损伤的保护作用,并观察对小鼠抗氧化应激的影响,旨在探讨人参皂苷干预AFL小鼠肝损伤的作用机制。方法:将60只清洁级健康SPF雄性小鼠,随机分为空白组、模型组、联苯双酯组和人参皂苷大、中、小剂量组,除空白组,其余组给予高脂饲料和酒精建立AFL模型,联苯双酯组给予0.9 mg/kg联苯双酯滴丸灌胃,人参皂苷大、中、小剂量组给予人参皂苷灌胃,剂量分别为15 mg/kg、30 mg/kg、60 mg/kg,空白组与模型组给予同体积的生理盐水灌胃。给药4周后,观察小鼠的体质量、肝脏质量、肝脏系数、中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDLC)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、血清瘦素(Leptin)、肿瘤坏死因子(TNF-α)、超氧化物歧化酶(SOD)、血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、全血抗氧化能力(T-AOC)、丙二醛(MDA)水平。结果:1各组的体质量差异无统计学意义(P>0.05);2与空白组对比,模型组的肝质量、肝脏系数,血清中TC、TG、LDL-C、ALT、AST、Leptin、TNF-α和肝脏MDA明显升高(P<0.01),血清中HDL-C、SOD、GSH-Px、T–AOC水平降低(P<0.01);3与模型组对比,人参皂苷各剂量组的肝脏系数、TC、TG、LDL-C、Leptin、TNF-α均明显下降(P<0.05或P<0.01);人参皂苷小剂量组ALT、AST水平降低(P<0.01);人参皂苷大剂量组和中剂量组血清SOD、血清GSH-Px水平升高,肝脏MDA水平降低(P<0.01);4与联苯双酯组对比,人参皂苷各剂量组LDL-C水平降低(P<0.01),AST较高(P<0.05或P<0.01);人参皂苷大剂量组小鼠血清Leptin、TNF-α水平降低(P<0.01)。结论:人参皂苷对AFL小鼠肝损伤具有保护作用,可能与人参皂苷具有抗氧化、改善血脂和肝功能、抗氧化应激、降低血清TNF-α和Leptin水平有关。Objective: To observe the protective effects of liver injury in alcoholic fatty liver mice by ginsenoside and observe the influence of oxidation stress in mice and mechanism of action.Method:Sixty clean grade healthy SPF male mice were randomly divided into blank group,model group,bifendate group and ginsenoside large,medium and small doses groups.Except blank group,the rest groups were given a high-fat forage and alcohol to set up model of AFL.The bifendate group was given 0.9 mg/kg bifendate and ginsenoside large,medium and small doses groups were given ginsenoside.The doses were 15 mg/kg,30 mg/kg,60 mg/kg,respectively.The blank group and model group were given the same volume of normal saline.Four weeks after treatment,the body weight,quality of the liver,liver coefficient,total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),alanine aminotransferase(ALT),aspertate aminotransferase(AST),blood thin element(Leptin),tumor necrosis factor(TNF alpha),superoxide dismutase(SOD),serum superoxide dismutase(SOD),glutathione peroxidase(gsh-px),whole blood antioxidant capacity(T-AOC) and malondialdehyde(MDA) in mice were studied.Result:(1)There was no statistically significant difference between groups for weights of mice(P〉0.05).(2)Compared with blank group,the quality of liver,liver coefficient,serum TC,TG,LDL-C,ALT,AST,Leptin,TNF alpha and liver MDA in model group increased significantly(P〈0.01).The serum HDL-C,SOD,GSH-Px and T-AOC were reduced(P〈0.01).(3) Compared with model group,the liver coefficient,TC,TG,LDL-C,Leptin and TNF alpha in each dose group of ginsenosides were significantly decreased(P〈0.05 or P〈0.01).Levels of ALT and AST in ginsenosides low group were lower(P〈0.01) and levels of serum SOD and GSH-Px serum in ginsenosides high-dose group and middle dose group were elevated and level of liver MDA decreased(P〈0.01).(4) Compa
