摘 要:目的探讨镇肝熄风汤抑制帕金森病肝阳上亢证大鼠脑组织氧化应激的可能机制。方法 105只雄性Wistar大鼠随机分为空白对照组、假手术组、模型组、镇肝熄风汤高、中、低剂量组[32,16,8 g/(kg·d)]及司来吉兰[0.9 mg/(kg·d)]组,每组15只。以附子汤灌胃联合6-羟基多巴胺单侧损毁黑质制备帕金森病肝阳上亢模型。镇肝熄风汤组大鼠按体重剂量灌胃,每日1次,共4周。分光光度法检测大鼠中脑组织MDA含量,Western blot检测海马Nrf2和HO-1蛋白表达。结果与假手术组大鼠比较,模型组大鼠中脑组织MDA含量明显升高,HO-1蛋白表达代偿性上调。与模型组比较,镇肝熄风汤显著降低了PD肝阳上亢证大鼠中脑组织MDA含量,上调Nrf2和HO-1蛋白表达。结论镇肝熄风汤对帕金森病肝阳上亢证大鼠中脑组织氧化损伤具有保护作用,其机制可能与镇肝熄风汤活化脑组织Nrf2抗氧化防御体系有关。Objective The aim of this study was to investigate the effects of Zhenganxifeng decoction on oxidative stress in the midbrain of Parkinson's disease rats with syndrome of hyperactivity of liver - Yang, and some mechanisms involved. Methods A total of 105 pathogen - free male Wistar rats were randomly divided to produce 7 groups : the normal control, sham - operated group, model group, Zhenganxifengdecoction group with three different dose (32, 16, or 8 g/kg) group, and selegiline group. Rat model of Parkinson's disease with syndrome of hyperactivity of liver - YANG was prepared by 6 - OHDA intra - substantia nigra injection and Fuzi decoction intragastric administration. Different doses of Zhenganxifeng decoction (32, 16, or 8 g/kg) were given via intragastric administration for 4 weeks. Measurements of MDA content were performed using spectrophotometry. Western blot were used to analyze the Nrf2 and HO - 1 protein expression. Results Zhenganxifeng decoction significantly inhibi- ted MDA content, upregulates the expression of Nrf2 and HO- 1 at level of translation in the midbrain of Parkinson's disease rats with syndrome of hyperactivity of liver - Yang. Conclusion Antioxidant properties of Zhenganxifeng decoction may be attributable to activation of Nrf2 antioxidative defense system in the midbrain of Parkinson's disease rats with syndrome of hyperactivity of liver - Yang.
