摘  要:目的研究槲皮素、放射线单独及联用对体外培养宫颈癌Hela细胞增殖、凋亡、迁移和侵袭的影响并探讨分子机制。方法槲皮素、放射线单独及联合作用Hela细胞后,MTT法检测细胞增殖,流式细胞法检测细胞凋亡,划痕法及Transwell法检测细胞迁移及侵袭,免疫印迹法检测MMP-9蛋白。结果槲皮素、放射线单独或合用均可对Hela细胞产生明显的抑制增殖和诱导凋亡作用(P<0.05),联用效果更好;放射线可刺激Hela细胞迁移及侵袭并上调MMP-9蛋白表达,而槲皮素则可抑制这种作用并下调MMP-9蛋白表达。结论槲皮素可通过诱导凋亡方式增强宫颈癌细胞对放射线敏感性,并可通过下调MMP-9蛋白表达抑制放疗期间癌细胞的侵袭和转移。Objective To study the effects of Quercetin, X - ray alone or combination on biological behaviour of Uterine Cervical Carcinoma Cell Line Hela cultured in vitro and explore its mechanism. Methods The growth and proliferation of Hela cell was detected by MTT assay . Cell apoptosis was detected by the flow cytometry. Cell invasive and migration ability was measured with Transwell assay and scratch adhesion test, respectively. The expression of MMP - 9 Protein was detected by Western blot. Results Quercetin, X -ray alone or combination all could inhibit cell proliferation and induce cell apoptosis( P 〈 0.05 ) , the result of combination was better than Quercetin,X - ray alone ; X - ray could stimulate Hela cell invasive and migration and up - regulate the expression of MMP - 9 Protein, while Quercetin could inhibit these effects and markedly suppress MMP - 9 Protein.. Conclusion Quercetin could enhance the sensibility to X - ray by inducing apoptosis and inhibit cell invasive and migration during radiation by down -regulating MMP- 9 in Hela cell cultured in vitro.
