黄连-吴茱萸药对中3种生物碱的高效液相色谱法同时测定

摘　　要：目的 建立同时测定黄连-吴茱萸药对中盐酸小檗碱、吴茱萸碱和吴茱萸次碱的方法。方法 采用HPLC法,色谱柱规格为Kromasil 100-5 C18（250mm×4.6mm,粒度5μm）;流动相：甲醇-乙腈-十二烷基磺酸钠（SDS）5 mmol·L^-1水溶液（2∶4∶4,p H 7.2）;流速1 ml·min^-1,柱温35℃,检测波长225 nm。结果 盐酸小檗碱、吴茱萸碱和吴茱萸次碱的线性范围分别为0.4004～5.005、0.4324～5.405、0.2168～2.171μg,平均加样回收率分别为：99.01%（RSD2.22%）、99.37%（RSD1.94%）、98.68%（RSD1.64%）。结论 此法操作简单,灵敏度高,可在225 nm处同时测定黄连-吴茱萸药对中3个有效成分的含量。Objective To develop an method for determining berberine hydroch]oride, evodiamine, and rutecarpine at the same time. Methods By using HPLC method, the chromatographic column was a Kromasil 100 -5 C18 column （250 mm × 4.60 mm, 5 μm ）. The mobile phase was methanol - aeetonitrile - SDS solution（5mmol·L^-1） （ 2：4：4, pH 7.2 ）. The flow rate was 1 mL ·min^-1. The column temperature was 35℃. The examining wavelength was 225 nm. Results The linear range of berberine hydroehloride, evodiamine, and ruteearpine was respectively 0. 4004 - 5. 005,0. 4324 - 5. 405,0.2168 - 2.171 μg. The average recovery of the added index was respectively 99.01% （RSD 2.22% ） ,99.37% （RSD 1.94% ） ,98.68% （RSD 1.64% ）. Conclusion The HPLC method was simple to use and sensitive. It can determine the content of three active ingredients in the couplet medicines of Coptidis and Medicinal Evodia Fruit at 225 nm.
