摘 要:目的从链霉菌中寻找抗HIV活性物质。方法对筛选得到的活性菌株进行菌种鉴定,采用大孔树脂结合制备的方法获得活性化合物,并对化合物进行了结构鉴定。结果菌株HCCB 10327初步鉴定为生二素链霉菌,活性物质经分离纯化和结构鉴定为siamycin Ⅱ和一个新的siamycin Ⅱ衍生物,这两个物质都具有较强的抗HIV活性,IC50分别为12.6和36.8μg/m L。结论首次从生二素链霉菌中发现了siamycin Ⅱ及一个新的衍生物。Objective To discover anti-HIV compounds from metabolites of Streptomyces. Methods The screened active strain was taxonomic identified. The active compounds were isolated with macro-porous resins and reversed-phase high performance liquid chromatography from the strain. Then, the structures of these compounds were elucidated through amino acid, MS and NMR analysis. Results The strain HCCB 10327 was identified as Streptomyces ambofaciens. Siamycin Ⅱ and a novel derivative were isolated and identified on the basis of amino acid and spectroscopic data analysis. Two compounds showed strong anti-HIV activities with IC50 of 12.6 and 36.8μg/mL. Conclusion Siamycin Ⅱ and its novel derivative were discovered from Streptomyces ambofaciens.
