摘 要:目的考察高效液相色谱法(HPLcl测万古霉素血药浓度时,以牛血清、人血清和人血浆作为空白基质制备标准曲线对万古霉素血药浓度的监测是否存在差异。方法采用AgilentTC-C18型色谱柱(4.6mm×150mm,5μn),甲醇.0.2%-乙胺缓冲液(磷酸调至pH3.2)为流动相,流速1.2mL/min,检测波长236nm,去甲万古霉素作为内标。结果3种基质中的万古霉素在3.41-109.0mg/L浓度范围内线性关系均良好,牛血清和人血清作为空白基质制备标准曲线的斜率差异较大,人血清和人血浆作为空白基质制备标准曲线的斜率差异不大。万古霉素和去甲万古霉素在3种基质中的提取回收率均存在较大差异。结论以牛血清、人血清和人血浆作为空白基质制备标准曲线对HPLC法万古霉素血药浓度的监测存在差异。Objective To study the differences among newborn calf serum, human serum and human plasma as blank matrixes when measuring vancomycin blood concentrations by high performance liquid chromatography (HPLC). Methods The detection of vancomycin blood concentrations by HPLC was performed on Agilent TC- Cls (4.6mmx150mm, 5μm) using a mobile phase consisting of methanol-0.2% triethylamine buffer (phosphoric acid adjusted pH=3.2) at the flow rate of 1.2mL/min. The detection wavelength was 236nm, and norvancomycin was used as internal standard. Results Vancomycin concentrations were in excellent linearity in the range of 3.41-109.0mg/L. The slopes of the standard curves were quite different between newborn calf serum and human serum as blank matrix, but the slopes of the standard curves were slightly different between human serum and human plasma as the blank matrix. And the extraction recoveries of vancomycin and norvancomycin were quite different in all three kinds of matrixes. Conclusion There were differences among newborn calf serum, human serum and human plasma as blank matrixes to prepare standard curves when monitoring blood concentration of vancomycin using HPLC method.
