摘  要:目的探讨重组人促红细胞生成素(rh EPO)对支气管肺发育不良(BPD)模型肺组织中酪氨酸激酶-2(JAK2)、信号转导和转录活化因子-5(STAT5)蛋白和mRNA表达的影响。方法将48只孕15 d的健康SD大鼠分别孕囊内注射脂多糖(LPS)或生理盐水,待其自然分娩后将新生大鼠随机分为对照组、高氧组、rh EPO组。采用Western blot和RTPCR法检测肺组织中JAK2、STAT5蛋白和mRNA的表达。结果高氧组JAK2、STAT5蛋白和mRNA表达减少,rh EPO组JAK2、STAT5蛋白和mRNA表达增加,高氧暴露第1、7、14天末,各组间JAK2、STAT5蛋白和mRNA表达的差异有统计学意义(P<0.05)。结论高氧组肺组织凋亡显著增加,rh EPO组肺组织凋亡减少,rh EPO对BPD模型肺组织的保护机制可能与活化JAK2/STAT5通路,减轻肺组织凋亡相关。Objective To explore the role of Janus activating kinase-2/signal transducer and activator of transcription-5( JAK2/STAT5) pathway in recombinant human erythropoietin( rh EPO) protecting BPD. Methods Injected LPS or NS to 48 sprague-dawley( SD) rats,gestational sacs on 15 th day of gestation,the newborn rats were divided into three groups: control group,hyperoxia group,rh EPO group. To detect the expression of lung tissue JAK2 and STAT5 protein and mRNA by Western blot and RT-PCR. Results In the hyperoxia group JAK2 and STAT5 expression was decreased,while in the rh EPO group,JAK2 and STAT5 expression was increased. The differences between the two groups were statistically significant on 1st,7th and 14 th day of hyperoxia exposure( P〈0. 05). Conclusion Intrauterine inflammatory exposure combined with hyperoxia after birth can cause lung cell apoptosis increases.While rh EPO has certain control effects on BPD,possibly by reducing the pulmonary apoptosis. The protection mechanism may be associated with JAK2/STAT5 signaling pathway.
