摘  要:目的观察辛伐他汀对肺癌A549细胞的作用,并探讨可能的作用机制。方法用不同浓度辛伐他汀处理A549细胞,采用MTT法检测其对A549细胞的生长抑制情况;用瑞士吉姆萨染色观察辛伐他汀对A549细胞形态学的影响;流式细胞仪检测活性氧(Reactive oxygen species,ROS)的浓度;实时荧光定量PCR测定miR-21、Rho A和K-Ras mRNA的表达。结果辛伐他汀对A549细胞的IC50=23μmol·L-1;形态学观察其能促进A549细胞凋亡且促使细胞内ROS明显升高;miR-21、Rho A和K-Ras mRNA的表达上调。结论辛伐他汀有一定的体外抑制A549细胞的效应;可能是通过改变细胞内的氧化应激水平来发挥的抗肿瘤效应。OBJECTIVE To investigate the inhibitory effect and the anti - tumor mechanism of Simvastatin on A549 cell line in vitro. METHODS MTT assay was used to assess the inhibitory effects of Simvastatin on proliferatin of A549 cell line. Wright Giemsa staining was used to observe the effects of Simvastatin on A549 cell line. Flow cytometry was employed to measure ROS level. Real - time quantitative PCR was employed to analyze the mRNA expression levels of miR - 21, RhoA and K - Ras. RESULTS IC50 of Simv- astatin to A549 ceils was 23 μm. Evident apoptosis phenomenons in A549 cell line were observed after treated with different concentra- tions of Simvastatin in Wright Giemsa. ROS level increased obviously in Simvastatin - treated cells, miR -21, RhoA and K - Ras mRNA were up - regulated in Simvastatin - treated cells. CONCLUSION Simvastatin has the inhibitory effect of inhibition towarding A549 cell line, which might be caused by the changes of intracellular redox state.
