摘  要:目的采用HPLC法同时测定塞北紫堇药材中北美黄连碱和原阿片碱的含量。方法采用Agilent Zorbax Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-醋酸钠缓冲液(55∶45),流速1 m L·min^(-1),检测波长280 nm,柱温30℃。结果测定了塞北紫堇药材中2种生物碱类成分的含量,在线性范围内的线性关系良好(r2>0.9999),平均回收率为99.13%~100.01%,RSD=1.09%~1.18%(n=6)。结论所用方法操作简便、准确、稳定、分离效果好,适用于同时测定塞北紫堇药材中2种生物碱的含量。OBJECTIVE To develop an HPLC method for simultaneous determination of 2 kinds of alkaloids ( β - hydrastin, proto- pine) in Corydalis impatiens. METHODS Agilent Zorbax Eclipse XDB - C18 column (250 mm ×4.6 mm, 5 μm)was used. The mobile phase consisted of acetonitrile - sodium acetate buffer (55 : 45 ) with the flow rate of 1 mL·min^-1, detection wavelength of 280 nm and column temperature of 30 ℃. RESULTS The determined contents of 2 kinds of alkaloids in C. impatiens showed that the new method had a good linear relationship within the linear range (r^2 〉 0. 9999) and the average recovery rate was in the range of 99.13% to 100.01% with the RSD ranging from 1.09% to 1.18% (n = 6). CONCLUSION The method is simple, accurate,reproducible and suitable for simultaneous determination of the contents of 2 kinds of alkaloids in C. impatiens.
