摘 要:目的建立HPLC法同时测定菝葜Smilax china L.和金刚藤糖浆(菝葜)中4种成分的含有量。方法该药材和药物甲醇提取液的分析采用Diamonil C18色谱柱(250 mm×4.6 mm,5μm);流动相乙腈-0.1%磷酸,梯度洗脱;体积流量1.0 m L/min;柱温30℃;进样量10μL;检测波长290 nm(落新妇苷)、306 nm(白藜芦醇)、360 nm(芦丁和山柰酚)。结果落新妇苷、白藜芦醇、芦丁、山柰酚分别在0.006 64~0.664μg(r=0.999 7)、0.006 58~0.658μg(r=0.999 9)、0.001 01~1.01μg(r=0.999 6)、0.0078~0.78μg(r=0.9999)范围内呈良好的线性关系,金刚藤中其平均加样回收率分别为98.40%、100.89%、99.11%、100.17%,RSD分别为2.1%、3.2%、2.1%、2.8%;金刚藤糖浆中其平均加样回收率分别为101.70%、97.50%、101.55%、101.23%,RSD分别为2.2%、1.3%、1.4%、1.4%。4种成分含有量在各批药材和制剂中均有明显差异,仅白藜芦醇能在所有样品中被检出。结论白藜芦醇可作为菝葜和金刚藤糖浆质量评价的指标成分。AIM To establish an HPLC method for the simultaneous content determination of four constituents in Smilax china L. and Jingangteng Syrup( S. china). METHODS The analyses of methanol extracts of the medicinal material and drug were performed on a 30 ℃ thermostatic Diamonil C18column( 4. 6 mm × 150 mm,5 μm),with the mobile phase comprising of acetonitrile-0. 1% phosphoric acid flowing at 1. 0 m L / min in a gradient elution manner,and the detection wavelengths were set at 290 nm for astilbin,306 nm for resveratrol,and 360 mm for rutin and kaempferol. RESULTS Astilbin,resveratrol,rutin and kaempferol showed good linear relationships within the ranges of 0. 006 64- 0. 664 μg( r = 0. 999 7),0. 006 58- 0. 658 μg( r = 0. 999 9),0. 001 01- 1. 01 μg( r =0. 999 6) and 0. 0078- 0. 78 μg( r = 0. 9999). The average recoveries of them in S. china were 98. 40%,100. 89%,99. 11% and 100. 17% with the RSDs of 2. 1%,3. 2%,2. 1% and 2. 8%,those in Jingangteng Syrup were 101. 70%,97. 50%,101. 55% and 101. 23% with the RSDs of 2. 2%,1. 3%,1. 4% and 1. 4%,respectively.The contents of four constituents were found to be obviously different among various batches of medicinal materials and preparations,and only resveratrol could be detected in all the samples. CONCLUSION Resveratrol can be taken as the indicative constituent in the quality control of S. china and Jingangteng Syrup.
