栀黄止痛贴质量标准的研究

摘　　要：目的建立栀黄止痛贴（栀子、大黄、黄柏等）的质量标准。方法 TLC法对栀子、大黄、黄柏、赤芍进行鉴别,HPLC法同时测定栀子苷、芍药苷、盐酸小檗碱、大黄酸的含有量,再对栀子苷释放度进行测定。结果 TLC斑点清晰,分离度良好,阴性无干扰。栀子苷、芍药苷、盐酸小檗碱、大黄酸分别在1.2~7.2μg（r=0.999 9）、4.32~25.92μg（r=0.999 8）、1.344~8.064μg（r=0.999 9）、1.664~9.984μg（r=0.997 0）范围内线性关系良好,平均加样回收率分别为99.40%（RSD=2.57%）、98.20%（RSD=2.25%）、97.45%（RSD=1.25%）、97.38%（RSD=1.51%）。栀子苷平均释放度为96.49%。结论该方法简便、准确、可靠,可用于栀黄止痛贴的质量控制。AIM To establish the quality standard for Zhihuang Zhitong Emplastrum（ for pain management,containing Gardeniae Fructus,Rhei Radix et Rhizoma,Phellodendri Chinensis Cortex,etc.）. METHODS Gardeniae Fructus,Rhei Radix et Rhizoma,Phellodendri Chinensis Cortex and Paeoniae Radix Rubra were identified by TLC. HPLC was adopted in the simultaneous content determination of gardenoside,paeoniflorin,berberine hydro-chloride and rhein,after which the release rate of paeoniflorin was investigated. RESULTS Free from negative interference,TLC spots were clear and well-separated. Gardenoside,paeoniflorin,berberine hydrochloride and rhein showed good linear relationships within the ranges of 1. 2- 7. 2 μg（ r = 0. 999 9）,4. 32- 25. 92 μg（ r =0. 999 8）,1. 344- 8. 064 μg（ r = 0. 999 9） and 1. 664- 9. 984 μg（ r = 0. 999 7）,whose average recoveries were 99. 40%（ RSD = 2. 57%）,98. 20%（ RSD = 2. 25%）,97. 45%（ RSD = 1. 25%） and 97. 38%（ RSD =1. 51%）,respectively. The average release rate of paeoniflorin was 96. 49%. CONCLUSION This simple,accurate and reliable method can be used for the quality control of Zhihuang Zhitong Emplastrum.
