摘 要:【目的】比较不同培养条件下滨梅幼苗耐盐特性差异,并筛选出耐盐能力强的滨梅品种,从而为滨梅耐盐性评价提供依据。【方法】以不同种源滨梅组培试管苗为试验材料,分别在组培和水培条件下,以0,100,200mmol/L NaCl进行盐胁迫处理,通过观察测定不同种源滨梅叶片盐害及生理生化指标,分析不同培养条件下滨梅幼苗对NaCl胁迫的响应情况。【结果】盐胁迫7d后,组培和水培滨梅幼苗叶片中Na+、丙二醛(MDA)、可溶性蛋白(SP)含量、相对电导率(REC)及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)活性均随NaCl处理浓度的升高显著增加;5种不同来源滨梅幼苗材料盐胁迫生理指标差异显著,其耐盐性强度表现为MI<MA1<NY<NJ<MA2,其中MA2可耐受200mmol/L NaCl,MI可耐受100mmol/L NaCl;组培条件下40d和水培条件下15d后,200mmol/L NaCl胁迫下NY、NJ、MA2所受盐害强度达到3级,MI、MA1所受盐害强度达到4级。【结论】不同浓度盐胁迫7d后组培和水培滨梅幼苗耐盐生理指标变化规律一致,滨梅组培试管苗可直接用于耐盐生理指标的测定和耐盐性评价。[Objective] This research investigated the influence of salt stress on physiological character- istics of Prunus maritima Marshall seedlings under different culturing conditions and selected new variety with strong salt tolerance. [Method] Five provenances of beach plum were treated with 0,100, and 200 mmol/L NaC1 under tissue and water culturing conditions to evaluate their salt-tolerance. [Result] With the increase of NaC1 concentration, MDA content, Na+ content, total soluble protein content (SP), relative conductivity,and activities of superoxide dismutase (SOD),catalase (CAT) and peroxidase (POD) in leaves of seedlings were significantly increased under both tissue and water culturing conditions 7 days after treatment. The physiological indexes of the five provenances were significantly different with the salt-tolerance in order of MI〈MAI〈NY〈NJ〈MA2,in which MA2 had salt-tolerance of 200 mmol/L NaC1 and MI of 100 mmol/L NaC1. After 40 days treatment under tissue culturing and 15 days treatment under water culturing,200 mmol/L NaCI caused class HI salt injury to NY,NJ,and MA2,and class IV to MI and MA1. [Conclusion] Salt injury and physicochemical indexes changed similarly under different culturing conditions,indicating that the tube seedlings of beach plum can be used to investigate the physicochemical inde- xes for salt-tolerance evaluation.
