摘 要:目的建立一种应用基因芯片技术检测临床病原微生物铜绿假单胞菌菌种及其耐药基因的方法,并进行鉴定。方法根据生物信息学设计引物及探针,采用基因芯片技术对分离51株临床病原微生物及其耐药基因进行检测,并以PCR结果做对照来评价芯片技术的特异性及灵敏度。结果采用基因芯片技术共检出41株铜绿假单胞菌,病原菌阳性检出率100.0%;耐药基因检测结果显示,有11株TEM基因阳性,15株VIM-2基因阳性,8株IntⅠ基因阳性,1株IMP-1阳性,4株阴性,其中39份基因芯片检测结果与PCR结果一致,准确率为95.1%。结论基因芯片系统具有高通量、高特异性等优点,对鉴定临床多药耐药铜绿假单胞菌有很大的应用价值,尤其能够为临床快速选择合适的抗菌药物提供很好的指导依据。OBJECTIVE To establish an application of gene chip technology to detect and identify the types of Pseudomonas aeruginosa and resistant genes of it.METHODS The primers and probes were designed by bioinformatics software.A total of 51 strains of pathogens isolated and the resistant genes were under detection of gene chip technology,and the PCR results were compared to evaluate the specificity and sensitivity of chip technology.RESULTS A total of 41 strains of P.aeruginosa were isolated by the way of gene chip technology,and the positive detection rate of target bacteria was 100%.The results of drug-resistant genetic testing showed that 11 strains of TEM genes were positive,15 strains of VIM-2genes positive,8strains of IntⅠ genes positive,1strain of IMP-1gene positive,and 4strains negative.The detection results of 39 gene chips were consistent with PCR and the correct rate was 95.1%.CONCLUSIONThe gene chip method has the advantages of high flux and high specificity,which could provide a huge application value for identify the multiple resistant P.aeruginosa,especially which could select the appropriate clinical antimicrobial and provide good guidance.
