摘　　要：目的研究脂氧素A4对脂多糖(LPS)诱导的人滋养细胞炎症细胞因子的影响及其可能的机制,为临床应用脂氧素A4治疗炎症相关性病理妊娠提供理论依据。方法分离纯化并鉴定人滋养细胞,分为对照组、LPS组、干预组和拮抗组,酶联免疫法检测细胞培养上清中TNF-α、IL-6、IL-10浓度,免疫印迹法检测细胞总蛋白中IκBα含量。结果与对照组相比,LPS组TNF-α、IL-6、IL-10浓度显著增加,IκBα含量显著减少;与LPS组相比,干预组TNF-α、IL-6显著减少,IL-10、IκBα显著增加;拮抗组TNF-α、IL-6高于干预组、但低于LPS组,拮抗组IL-10、IκBα低于干预组、但高于LPS组。结论脂氧素A4下调LPS诱导的促炎细胞因子表达、上调抑炎细胞因子表达,促进炎症消退,这种效应可能通过影响NF-κB的活化来实现。OBJECTIVE To investigate the effects of lipoxin A4 on inflammatory cytokines induced by lipopolysaccharide（LPS）in human trophoblast cells and its possible mechanism,so as to provide theory reference for lipoxin A4 on inflammatory relative pathologic gestation.METHODS Primary human trophoblast cells were isolated,purified and detected,which were randomized into four groups：control group,LPS group,intervention group,and antagonistic group.The concentration of TNF-α,IL-6and IL-10 in the supernatants were analyzed by ELISA and the level of IκBαprotein were observed by Western Blot.RESULTS Compared with control group,the concentration of TNF-α,IL-6and IL-10 in LPS group were significantly improved,the level of IκBαprotein significently decreased.Compared with LPS group,the concentration of TNF-αand IL-6in intervention group were significantly lower,the concentration of IL-10 were significantly higher,the level of IκBαprotein significently increased.The concentration of TNF-αand IL-6in antagonistic group were higher than intervention group,but lower than LPS group,the concentration of IL-10 and the level of IκBαprotein in antagonistic group were lower than intervention group,but higher than LPS group.CONCLUSIONLipoxin A4down-regulates pro-inflammatory cytokines and upregulates anti-inflammatory cytokines induced by LPS.These effects may be associated with NF-κB activation.
