摘 要:目的对2013年云南省22株柯萨奇病毒A6型(coxsackievirus A6,CV-A6)VP1区基因特征进行分析。方法对云南省2013年手足口病实验室监测系统分离到的22株CV-A6病毒VP1区基因进行基因扩增和测序;按参考文献下载基因库中CV-A6病毒VP1区基因参考序列,用MEGA 5.0软件计算不同毒株的核苷酸和氨基酸差异率,并构建系统进化树,进行基因特征和分子流行病学分析。结果 2013年云南省在711份非肠道病毒71型(EV-A71)、非柯萨奇病毒A16型(CV-A16)肠道病毒中共检出CV-A6病毒22份,占3.09%(22/711),CV-A6的阳性总检出率为0.32%(22/6 912)。基因特征分析表明,云南22株CV-A6均为基因1型(Genotype 1)。它们分为3个不同的进化分支,1株为China cluster,6株为Yunnan cluster 1,15株为Yunnan cluster 2。结论除原型株Gdula(AF081297)外,国内外CV-A6病毒存在两个基因型,基因型2又分为8个进化分支,云南分离株分布在3个不同的进化分支,2013年云南省CV-A6的流行存在3个传播链。Objective To analyze the genetic characteristics of coxsackievirus A6(CV-A6)VP1 gene sequences isolated from hand,foot and mouth disease(HFMD)from Yunnan province of China in 2013. MethodsRT-PCR and VP1 gene sequencing were done for 22 strains of CV-A6 isolated from HFMD surveillance system in 2013.VP1 gene sequences of CV-A6 reference strains were downloaded from the GenBank and their nucleotide(nt)and amino acid(aa)diversities were calculated by MEGA 5.0 software,the phylogenetic tree was constructed and the genetic characteristics and molecular epidemiology were analyzed. Results In 2013,a total of 22 strains of CV-A6 viruses were detected from 711non-EV-A71 and non-CV-A16 EVs,accounting for 3.09%(22/711);the overall positive rate was 0.32%(22/912).Phylogenetic analysis showed that all 22 Yunnan CV-A6 strains were genotype 1and were further divided into 3 genetic clusters.One strain was grouped to China cluster,6strains to Yunnan cluster 1and 15 strains to Yunnan cluster 2. Conclusions The CV-A6 strains isolated in Yunnan province of China in 2013 can be genetically categorized into two genotypes(genotype 1and 2),genotype 2 can be further divided into eight clusters,Yunnan strains were distributed in 3 clusters,indicating there were 3CV-A6 transmission chains.
