细胞毒性试验中两种细胞计数方法的比较

摘　　要：目的比较Countstar自动细胞计数仪法和传统显微镜计数法两种计数方法在细胞毒性试验中细胞计数的差异。方法选用常见的两种类型的药品稀释液,稀释液为真溶液的注射用左旋泮托拉唑钠和稀释液为混悬液的毛蚶提取物,设立不同剂量组,细胞染毒6 h后制备细胞悬液,台盼蓝染色,分别用显微镜和Countstar自动细胞计数仪计数,对结果进行比较分析。结果 Countstar自动细胞计数仪法和显微镜计数法两种计数方法相关性良好,细胞计数结果差异无统计学意义(P>0.05)。结论 Countstar自动细胞计数仪法是相对简单、快捷、准确的细胞计数方法。Objective To compare the difference of two kinds of counting methods in cytotoxicity test,Countstar automatic cell counting method and traditional microscope counting method. Methods Selection of two types of common drug dilution,diluent for the true solution by Left- handed pantoprazole sodium and diluent for the suspension by the extract of Scapharca subcrenata,both of them were set different doses,cell suspension was prepared and stained by trypan blue after cells were exposed at 6 h,then we count with manual microscopic counting and Countstar automatic cell counting respectively. The results were compared and analyzed. Results Under the condition of this study,Countstar automatic cell counting method and the traditional microscope counting method showed a good relarivity,no significant diference was found（ P〈0. 05）. Conclusion With the two cell count methods,Countstar automatic cell counting is relativity simple,fast and accurate cell counting method.
