摘  要:促微管聚合蛋白(tubulin polymerization promoting proteins,TPPPs)家族是一类能与微管蛋白结合,从而促使微管聚集、维持微管系统稳定性和完整性的蛋白质,其在细胞有丝分裂纺锤体组装中发挥重要作用。目前已知该家族在人类基因组中有3个成员:TPPP/p25、TPPP2/p18和TPPP3/p20,其共同点是在C末端都包含一个保守的p25-α序列。TPPP1主要与神经系统疾病相关,如多发性硬化和多系统萎缩;TPPP3在许多肿瘤细胞中表达,与肿瘤的增殖密切相关;而TPPP2的研究相对较少。本文将主要阐述TPPP家族成员及其相关疾病的最新研究进展。Tubulin polymerization promoting proteins (TPPPs) can promote tubulin polymerization into microtubules and induce their bundling. They also function as the stabilization of microtubular network and play a crucial role in mitotic spindle formation. There are TPPP paralogs in the human genome: TPPP/p25, TPPP2/p18 and TPPP3/p20. The common C-terminal part of the 3 proteins is named as p25-α domain. TPPP1 is involved in the neurodegenerative disorders, such as multiple sclerosis and multiple system atrophy. TPPP3 is expressed in some tumor cells, and closely related to tumor cell proliferation, while TPPP2 is almost unknown so far. In this review, TPPPs and its related diseases is discussed in detail.
