摘 要:目的探讨新诊断T2DM患者与健康人群血清microRNA(miRNA)表达谱的差异表达。分析利拉鲁肽治疗前后血清miRNA-19a表达水平的变化,并分析其与胰岛β细胞功能变化的相关性。方法采用μParaflo^(TM)miRNA微流体芯片技术检测并筛选出血清中表达丰度有变化的miR-19a,分别测定使用利拉鲁肽治疗前后HbA_1C、血清miR-19a表达和胰岛功能水平。结果芯片检测发现,差异表达最明显的miRNA有12个,选取miR-19a进行PCR验证。利拉鲁肽治疗12周后,血清中miR-19a表达量和HbA_1c分别为(0.04±0.03)%、(6.13±0.57)%,较治疗前降低;胰岛素0~30 min曲线下面积(AUC_(Ins 0~30))与血糖AUC(AUC_(g 0~30))比值(INSR30),AUC_(Ins 0~120)/AUC_(g 0~120)比值(INSR120)及Matsuda胰岛素、敏感指数分别为(1.93±0.63)、(3.94±2.27)及(7.86±3.81),较治疗前增加(P<0.05)。结论利拉鲁肽治疗新诊断T2DM,胰岛β细胞功能的改善可能与血清中miR-19a表达下调有关。Objective To explore the differences of serum miRNA expression between patients with newly diagnosed type 2 diabetes mellitus (T2DM) and healthy people; analyze the expression of miRNA- 19a before and after Liraglutide treatment, and to analyze the association between miRNA-19a andpancreatic β cells' function. Methods MiR-19a was screened and tested by ParafloTM miRNA microfluid chip technology. The level of HbAlc, miRNA-19a and pancreatic function were measured in newly diagnosed T2DM patients before and after 12-weeks' Liraglutide treatment. Results A total of 12 miRNAs with the most significant differential expression were found in both newly diagnosed T2DM patients and healthy people, miR-19a was validated by PCR. After 12 weeks of treatment, the expression of miRNA-19a and HbAlc were significantly decreased (0.04±0.03)%、(6.13±0.57)%vs(7. 76±1.24)%, respectively, P 〈 0.05 ], whereas AUCIns o-30/AUCg 0-30, AUCIns 0-120/AUCg o-120 and matsuda index were signifieantly inereased(1.93±0.63)、(3.94±2.27)vs(7.86±3.81) respectively,P〈0. 05]. Conclusion In newly diagnosed T2DM treated with Liraglutide,the improvement of pancreatic islet beta cell function may be associated with the down- regulation of miR-19a expression.
