摘  要:高尔基体磷蛋白3(Golgi phosphoprotein 3,GOLPH3)在正常细胞中是一种磷酸化的高尔基体膜蛋白,参与执行高尔基体的蛋白质分选功能。GOLPH3已报道参与多种人类癌症,研究表明GOLPH3是一个全新的致癌基因,能促进肿瘤细胞的增殖和迁移,同时也可能是肿瘤治疗的新靶点。然而,其在胰腺癌中的临床意义和生物学作用尚不清楚。该研究通过重组慢病毒表达RNAi的方法,构建稳定细胞系使GOLPH3基因在胰腺癌细胞株colo357中表达沉默,同时构建过表达GOLPH3基因的colo357稳定细胞系,通过MTT实验,伤痕愈合实验以及Transwell实验研究GOLPH3基因在胰腺癌细胞中的增殖、侵袭以及转移能力中的作用。结果显示沉默GOLPH3基因的表达可抑制人胰腺癌细胞的增殖力、迁移力和侵袭力,提示GOLPH3有可能成为潜在的肿瘤标志物和独立的预后因子。Golgi phosphoprotein 3(GOLPH3) is a phosphorylated Golgi membrane protein in normal cells,which plays a role in anterograde and retrograde Golgi trafficking.GOLPH3 has been reported to be involved in the development of several human cancers.The studies show that GOLPH3 is a new oncogene,which can promote the proliferation and migration of tumor cells.It also can be used as a new therapeutic target.However,its clinical significance and biological role in pancreatic cancer remains unclear.In this study,we constructed a stable GOLPH3 silence colo357 cell lines,a pancreatic cancer cell line,using recombinant expression lentivirus RNAi method.GOLPH3 was also introduced into colo357 cell lines to construct a GOLPH3 overexpression line.To study the role of GOLPH3 in proliferation and metastasis of pancreatic cancer cells,MTT assays,scratch migration assays and Transwell experiments were used in this research.GOLPH3 silencing suppresses the capacities of proliferation,migration and invasion of human pancreatic cancer cells,suggesting that GOLPH3 may be a potential tumor marker and independent prognostic factor.
