二甲双胍对慢性髓性白血病细胞K-562增殖、凋亡及周期的影响

摘　　要：目的:研究二甲双胍(metformin,Met)对慢性髓性白血病(chronic myeloid leukemia,CML)细胞K-562增殖、凋亡、周期的影响及可能机制。方法:不同浓度的二甲双胍(终浓度0、0.1、0.3、1、3、10、30 mmol/L)对K-562细胞进行干预,CCK-8法检测细胞增殖,流式细胞术检测细胞凋亡及周期,Western blot法检测凋亡相关蛋白BCL2/BAX及周期相关蛋白cyclin D3/CDK6的表达。结果:二甲双胍不同时间点(12、24、36、48 h)不同浓度(终浓度1、3、10、30 mmol/L)均可显著抑制K-562细胞增殖,诱导K-562细胞凋亡同时使K-562细胞阻滞于G_0/G_1期;不同浓度(终浓度1、3、10、30mmol/L)二甲双胍作用K-562细胞后,凋亡相关蛋白BCL2/BAX的比值减小,周期相关蛋白cyclin D3、CDK6表达减少。结论:二甲双胍对K-562细胞有抑制增殖、促进凋亡以及阻滞周期的作用,其作用机制可能与其下调凋亡相关蛋白BCL2/BAX的比值,下调周期相关蛋白cyclin D3、CDK6的表达有关。AIM： To investigate the effect of metformin on proliferation, apoptosis and cell cycle of chronic myeloid leukemia cell line K-562. METHODS：CCK-8 assay was used to detect met formin effect on cell proliferation. Flow cytometry was used to analyze the cell apoptosis and cell cy- cle. The expression of apoptosis-related proteins BCL2/BAX and cell cycle-related proteins cyclin D3/CDK6 were determined by western blotting. RESULTS ： Metformin inhibited proliferation, induced apoptosis and induced significant increase of the G0/G1 phases fraction of K-562 cell. The ex-pression of cyclin D3 and CDK6 were down-regula- ted and the ratio of BCL2/BAX was decreased after metformin treatment. CONCLUSION： Mefformin can inhibit the proliferation, promote the apoptosis and induce the increase of the G0/G1 phases＇fraction of K-562 cell. The mechanism may be related to the down-regulation of cyclin D3, CDK6 and the ratio of BCL2/BAX.
